4 The effects of γ-secretase inhibitor DAPT on proliferation and apoptosis of U2OS
HU Guohai DAI Guo LIU Gaiwei SONG Qi YU Ling YANG Jian GUO Weichun
Objective To observe the effect of gamma secretase inhibitor DAPT on proliferation and apoptosis of human osteosarcoma cell line U2OS, and to explore the possible mechanism of DAPT inhibiting osteosarcoma proliferation and inducing apoptosis. Methods Osteosarcoma cell line U2OS was cultured in vitro. Cell proliferation was tested by cell counting kit-8 (CCK-8) assay. Double stained by Annexin V-FITC and Propidium Iodide (PI), the cells were detected by flow cytometry (FCM) for apoptosis. Hoechst 33258 was used to detect the morphological change of typical apoptotic cells. The apoptosis related mRNA and protein levels in U2OS were detected by RT-qPCR and Western blot. The activities of Caspase-3, Caspase-8 and Caspase-9 were measured with Caspase activity assay kit. Results CCK-8 assay showed that 5-50 μmol/L DAPT inhibited the proliferation of human osteosarcoma U2OS cells and in a time- and dose-dependent manners. Flow cytometry showed that 5, 10 and 20 μmol/L DAPT could significantly induce apoptosis of osteosarcoma U2OS cells, compared to the control group, the differences were statistically significant (P < 0.05). Apoptotic body could be observed by Hoechst 33258 staining after treatment with 5, 10 and 20 μmol/L DAPT. RT-PCR and Western blot analysis showed that after treatment with DAPT (5, 10, 30 μmol/L), the expression of Caspase-3, 8 ,9 mRNA were higher than those of control group (P < 0.05), the pro-apoptotic protein Bax increased (P < 0.05), and anti-apoptosis protein Bcl-2 protein decreased (P < 0.05). The activities of Caspase-3, Caspase-8 and Caspase-9 promoted obviously after treatment with DAPT (5, 10, 20, 30 μmol/L) when compared with the control group (P < 0.05). Conclusion DAPT can significantly inhibit the proliferation and induce apoptosis of human osteosarcoma cell line U2OS. Its mechanism of action may be related to activation of Caspase dependent apoptosis pathway.

MAO Xijing ZHU Bochi YU Tingmin YAO Gang▲
Objective To study the effects of 3-n-butylphthalide (NBP) on memory and Caspase-3 expression in the hippocampal CA1 region of vascular dementia rats, in order to explore protective effects and mechanisms of NBP on rats with vascular dementia (VD). Methods A total of 80 SPF Wistar rats were divided into vascular dementia models group (VD group), vascular dementia models + NBP injection group (NBP treatment group), sham surgery + NBP injection group (NBP control group), sham surgery group (Sham group), according to random group method. Then each group was divided into four subgroups (n = 5): 1, 2, 4, and 8 weeks after surgery. VD models were established by ligating bilateral common carotid artery. Then the rats in the NBP treatment group and NBP control group were intraperitoneally injected with NBP 5 mg/(kg·d) for 7 consecutive days. The rats in the VD group and Sham group were intraperitoneally injected with saline (0.2 mL/d) for 7 consecutive days. Memory ability of 8 weeks subgroup rats were tested by Morris water maze at 4 and 8 weeks after surgery. At 1, 2, 4, and 8 weeks after surgery, the rats in each group were decapitated. The brains were obtained, and then hippocampus were isolated. Caspase-3 expression in the hippocampal CA1 region were determined by immunohistochemistry. Results Compared with VD group, escape latency in the NBP treatment group were significantly shorter (P < 0.05). At 1, 2, 4 and 8 weeks after surgery, Caspase-3 expression in the VD group and NBP treatment group were significantly increased than those in the Sham group (P < 0.05). Compared with VD group, Caspase-3 expression in the NBP treatment group were significantly lower at 4 and 8 weeks after surgery (P < 0.05). In VD group, Caspase-3 expression at 8 weeks after surgery was significantly higher than at 1 weeks (P < 0.05). Conclusion NBP can improve the memory ability of VD rats, which may be connected with the overexpression of caspase-3 in the hippocampus CA1 region of VD rats.

93 Comparative study of the medical development history of Chinese and western "aromatherapy"
LI Zhiyue1 LI Feng1 ZHANG Yu1 SHAO Haozhen2 ZHANG Weiyue1 YU Jiaojiao1
"Aromatherapy" is originated from ancient civilizations - ancient Egypt and ancient China, which is originally used as customs and experiences in medicine; with the born of different historic cultures, philosophy and medicine in China and the West, Chinese and western "aromatherapy" becomes different: traditional Chinese medicine "aromatherapy" is classified into the category of external treatment of Chinese medicine - "aromatherapy therapy" and "fumigation therapy", it has been developed and matured in the aspects of drug type, dosage form, usage and theory, and has been used so far; western medicine "aromatherapy" has experienced the development and maturity of extracting essential oils, replacing essential oils by chemical synthesis, returning plant extracts and carrying out clinical research. There are such academic differences between Chinese medicine and western medicine "aromatherapy". In the final analysis, they are the differences of thinking patterns: the traditional Chinese medicine pays attention to the integrity of the body itself, and the unity and connection between human, nature and social environment; however, the reduction theory of western medicine divides the whole into several elements, and then makes a detailed analysis of the elements in order to trace the evidence and essence. To understand the similarity and difference of medical development paths between Chinese and western, and to seek common ground and fit points between them will bring about breakthroughs in the development and innovation of traditional Chinese medicine.
