Studies on the extracting and antioxidant activities of total flavonoids in Mongolian medicine Gymnadenia conopsea |

Abstract Objective To optimize the extraction process of total flavonoids in Gymnadenia conopsea, establish a method of its content determination and research its antioxidant activities. Methods The orthogonal experiments was used to optimize the extraction process of total flavonoids in Gymnadenia conopsea. The content of total flavonoids were determined using rutin as reference by colorimetric. The antioxidant activity was evaluated by hydroxyl free radical scavenging activity. Results The optimized extraction conditions were 90% ethanol as solvent, material -liquid ratio of 1∶20, reflux extraction 2 times, each time 60 min. Rutin showed good linear relationship in the range of 0.1094-0.8752 mg, the average recovery was 98.8%, RSD was 1.05%. The total flavonoids possessed potential scavenging activity of hydroxyl free radical. Conclusion The optimum extraction process is stable, reliable, the method of content determination is simple, accurate, reproducible. The content of total flavonoids is quite different in Gymnadenia conopsea of different areas. The total flavonoids in Gymnadenia conopsea have obvious antioxidant activities.
