FU Haiyang1 LI Min1,2 YU Hao1,2 FU Bufang1 WANG Zhaoxu1
Objective To study the changes of vascular structure in vitro stress culture of carotid artery in New Zealand rabbits by adjusting the parameters of fluid flow, flow rate and periodic pulsation. Methods Carotid arteries of SPF New Zealand rabbits were put in a bioreactor to establish the vessel culture model in vitro. There were four groups, the static group was static culture, and the dynamic groups (group 80, group 50 and group 25) were maintained under the same operating displacement and frequency of pulse pump, and the flow rates of culture medium were 80, 50 and 25 ml/min, respectively, to maintain the stress state culture. After the culture, the changes of vascular structure were evaluated by HE, Masson and EVG staining. Results In static group, vascular endothelial cells remained intact, smooth muscle cells were arranged in disorder, elastic fibers contracted sharply, and collagen fibers remained basically unchanged at 15 days. In group 80, elastic fibers did not change significantly, and endothelial cells could not be seen, and the smooth muscle in the 8th day was more orderly than that in the 4th day, and collagen fibers decreased significantly in the 8th day. In group 50, endothelial cells were not visible, smooth muscle cells were arranged in order, elastic fibers became loose and flat at the 15th days, and vascular collagen fibers decreased significantly. In group 25, the changes of elastic fibers and collagen fibers were milder than those in group 50. Conclusion Under the condition of simulating various parameters in vivo, the dynamic culture of blood vessels in vitro is helpful to maintain the physiological structure of blood vessels. The difficulty of culture is to prevent pollution and supplement collagen. Under the condition of high stress, vascular endothelial cells will be damaged, resulting in the proliferation of smooth muscle cells and the more serious loss of collagen fibers.

50 Research progress of FAM83A in malignant tumors#br#
ZHU Qiumin XU Min
FAM83A, namely the family with sequence similarity 83 member A gene, is a protein-coding gene containing domain 1669 whose function remains to be defined. Initially identified as a potential tumor gene, it is up-regulated in lung cancer, liver cancer, breast cancer, and other malignant tumors, promoting cancer cell proliferation, migration, invasion, epithelial-mesenchymal transformation, and other functions. Current studies suggest that a variety of microRNAs can act as tumor suppressor genes, and circRNA or long non-coding RNA is its upstream, directly or indirectly targeting FAM83A to form an axis to act on signaling pathways and inhibit the occurrence and development of cancer. However, the specific molecular mechanism remains to be further studied. This article reviews the expression and role of FAM83A in malignant tumors.

WANG Tong1* WANG Jinge2,3* JIA Chen1 HUANG Jingjing1 HUO Xiaoyan4 MENG Qinghui1 LIANG Desen1 PAN Huayang1
Colorectal cancer is one of the most common malignant tumors of the digestive tract. Its incidence and mortality are increasing year by year. Because its early symptoms are not obvious, it is mostly in the advanced stage when it is discovered, leading to poor prognosis and survival. Long non-coding RNA is an RNA molecule, which consists of more than 200 nucleotides in length and lacks protein-coding functions, but it has a crucial regulatory role at the level of epigenetic, transcription, post-transcriptional. Studies find that long non-coding RNA is closely related to the occurrence and development of colorectal cancer, the underlying mechanism mainly includes direct binding with RNA, DNA, and protein to play a biological role. Therefore, this article describes the research progress of long non-coding RNA in colorectal cancer, which can provide a theoretical basis and practical direction for a specific diagnosis, targeted therapy and prognostic evaluation of colorectal cancer.

TIAN Li ZHANG Shaoyan WU Xianwei QIU Lie LIU Xifang WU Dingzhong ZHANG Huiyong LU Zhenhui
Bronchiectasis is a chronic inflammatory bronchial disease caused by infection and bronchial obstruction. It is characterized by repeated attacks and long treatment course. Due to the increased incidence and mortality of the disease, it has gradually attracted worldwide attention. Previous studies in China mostly adopte endotracheal infusion of Pseudomonas aeruginosa to establish the model of bronchiectasis in rats. Foreign studies mostly use the mouse model of bronchiectasis. In recent years, organoid models have been added to provide more alternative models for basic research on bronchiectasis. In this review, we reviewed the progress of the mouse model of new bronchiectasis, including alginate, agar or agarose globules, endotracheal intubation with Pseudomonas aeruginosa, Mycobacterium abscess, and Stomatococcus mucilaginosus. The model of bronchiectasis is established in IFT88, Cby, Pkd2, and other ciliated gene knockout mice, and the model of respiratory tract organ such as human lung organ and gas-liquid interface tissue culture was established for reference in the study of this disease.

147 Experience summary on diagnosis and treatment of chronic gastritis by famous veteran traditional Chinese medicine doctor Cui Minlu#br#
WANG Xiongli1 LIN Mingyu1 LI Chunxiang2 CHEN Lifen3
Cui Minlu’s treatment of chronic gastritis is due to deficiency of qi and yang, stagnation of dampness and heat, and the combination of cold and heat, resulting in the disorder of ascending and descending, stomach disharmony and descending. Therefore, the treatment principle is to take care of the middle yang, transport spleen for eliminating dampness, relieve oppression and eliminate distension, and recover its rise and fall. Chronic gastritis is associated with psychological factors, from the point of view of five viscera storing spirit and spleen as pivot and ascending and descending pivot of spleen and stomach, traditional Chinese medicine should focus on regulating spleen and stomach, restoring its ascending and descending, supplemented with tranquilizing products. The recognition of chronic gastritis caused by Helicobacter pylori infection is more in line with the theory of dampness toxin in traditional Chinese medicine. In terms of treatment, the three factors should be taken into consideration in combination with the regional characteristics of Southern Fujian, in the first half of the year, the growth is more than enough, but in the second half of the year, the collection is insufficient. Spring and summer are long, autumn and winter are short, damp and hot are common, yang in constitution is insufficient, and spleen and kidney are deficient in the later period. Therefore, on the basis of clearing heat and removing dampness, we should pay attention to the care of zhongyang, it is advocated to warm and transport spleen yang to help clear away dampness and heat, combined with the progress of modern medicine, the micro syndrome differentiation, combined with endoscopic lesions, pay attention to the addition and subtraction of special drugs, pay attention to nourishing the kidney and latent yang in the later stage, pay attention to its root, recover its rise and fall, which is worthy of popularization and application.

GUO Huijuan1 CHEN Xun1 ZHANG Yinqiang2 GONG Yaoyao3 ZHENG Ying1 WANG Fengmei1 LI Qi1
Objective To analyze the results of the human immunodeficiency virus (HIV) antibody screening test and confirmation test, and to explore the reasons for false positives in the screening test. Methods From January 2017 to December 2019, a total of 184 HIV antibody screening-positive patients in Xiyuan Hospital, Chinese Academy of Traditional Chinese Medicine were sent to the confirmation laboratory for confirmation test, the test results were analyzed and statistics, and the characteristics and causes of false positive patients were explored. Results The false positive rate of HIV antibody screening increased year by year from 2017 to 2019, and the difference was statistically significant (P < 0.05). The screening test values of false-positive patients were concentrated in a relatively low interval, and the specimens with a screening value of 1≤S/CO<10 confirm that the verification was indeed all false positives; the false-positive rate decreased with the increase of S/CO value, and the specimens with S/CO≥100 were confirmed to be all positive. False positive patients have different basic medical histories. Among them, infection, cardiovascular and cerebrovascular diseases, autoimmune diseases, etc. interfered with the screening experiment to a certain extent, leading to the appearance of false positives. Conclusion For HIV antibody screening positive patients with relatively low S/CO, attention should be paid to the patient’s basic medical history and personal condition, whether there is interference in the test, and reasonable communication and explanation to the patient to reduce the patient’s psychological burden.
