YAN Li SHEN Dingzhu
Objective To explore the mechanism of Bushen Jiangzhi Formula in regulating autophagy and cholesterol efflux in the intervention of atherosclerosis (AS). Methods One hundred ApoE-/- mice induced by high fat diet were divided into model group, kidney tonifying group, statin group and 3-methyladenine (3-MA) group by using random number table method, with 25 mice in each group. Twenty-five C57BL/6J mice were normal controls. Hematoxylin-eosin (HE), oil red O and FILIPIN staining were used to detect the pathological morphology, lipid accumulation and free cholesterol (FC) staining of the aortic sinus. Autophagosomes and lipid droplets were observed by electron microscopy. The content of serum total cholesterol (TC) and FC were detected by colorimetry. The expression of liver X receptor alpha (LXRα), ATP-binding membrane cassette transport protein A1 (ABCA1), microtubule-associated protein 1 light chain 3 (LC3) and p62 protein were detected by Western blot. Results The model group had more aortic sinus AS plaque, lipid accumulation and FC positive staining area than the normal group, and the differences were highly statistically significant (P < 0.01). The normal group had intact aortic endothelial cell structure and autophagosome. The endothelial cells of aorta in model group were swollen with lipid droplets, but no autophagosomes were observed. The contents of TC, FC, and cholesterol ester (CE) in model group were higher than those in normal group, and the differences were highly statistically significant (P < 0.01). The expression of LXRα, ABCA1, and LC3 Ⅱ/Ⅰ in aorta of model group was lower than that of normal group, and the expression of p62 was higher than that of normal group, the differences were highly statistically significant (P < 0.01). AS plaque, lipid accumulation, and FC positive staining area of aortic sinus in the kidney tonifying group were less than those in the model group, and the differences were highly statistically significant (P < 0.01). In the kidney tonifying group, autophagosomes were found in the endothelial cells of aorta. The contents of TC, FC, and CE in kidney tonifying group were lower than those in model group, and the differences were statistically significant (P < 0.05 or P < 0.01). The expression of LXRα, ABCA1, LC3Ⅱ/Ⅰ in kidney tonifying group was higher than that in model group, while the expression of p62 was lower than that in model group, the differences were statistically significant (P < 0.05 or P < 0.01). Conclusion Bushen Jiangzhi Formula may effectively intervene in the treatment of AS by regulating autophagy and cholesterol efflux.

35 Research progress of autophagy in the prevention and treatment of malignant hematologic diseases#br#
WANG Wenru TANG Xudong
Autophagy can remove abnormal substances and organelles in cells in the early stage of malignant hematopathy, and help malignant cells to obtain various substances and energy in the later stage, and promote the growth and proliferation of tumor cells, which has a two-way effect. In clinical practice, chemoradiotherapy, demethylation, and targeted therapy can all activate autophagy, induce type Ⅱ programmed cell death of tumor cells and clear malignant cells, but a large amount of substances produced can help the survival of residual tumor cells and lead to drug resistance. It has been found that the combination of autophagy inhibitors can effectively improve the therapeutic effect of malignant hematologic diseases, especially for patients with relapsed and refractory diseases. In clinical treatment, how to effectively regulate the level of autophagy, remove tumor cells, and inhibit the progression of the disease are the key to the prevention and treatment of malignant hematologic diseases.

40 New advances in cognitive impairment associated with depressive disorder and its evaluation#br#
HAO Wensi1 WEI Min1 LIU Jianghong1 SONG Penghui1 WANG Jing2 WANG Yuping1
Depressive disorder is a common mental disorder characterized by emotional symptoms, such as low mood, slow thinking, and decreased will activity. More and more evidences indicate that the cognitive impairment in patients with depressive disorder is mainly related to the impairment of the frontal parietal cognitive control network. Of these, executive function, memory, and attention are the most commonly affected areas and can be assessed by specific neuropsychological tests. Executive function can be evaluated by trail making test part B, Stroop color-word test, etc. Memory is assessed by Wechsler memory scale and Rey auditory verbal learning test, and attention is assessed by continuous performance test, Delis-Kaplan executive function system test, CogitEx Ⅱ. At present, there are new comprehensive assessment methods for the cognitive function of patients with depressive disorder, such as THINC-it, severe cognitive impairment profile, cognitive component of rating bipolar disorder. A more detailed assessment of specific areas of cognitive impairment in patients with depressive disorders will improve the accuracy of screening and diagnosis, and have great implications for molecular studies and future targeted therapies.

53 Mechanism of pyroptosis and its role in various kidney diseases#br#
SUN Xiaoyi1 JIANG Yuqin1 TANG Yuyan2 LIU Hejing1 LU Xun3 WEI Minggang2
Pyroptosis is a newly discovered method of pro-inflammatory programmed cell death in recent years, which can be caused by inflammasome, caspase-1, human caspase-4/5, and mouse caspase- 11, and its downstream response factor gasdermin D (GSDMD) co-mediated. When the body is invaded by foreign substances, it can initiate the upstream reaction element of pyroptosis, which promotes the structural change of GSDMD. After more GSDMD N-terminal necrotic domains accumulate to form a membrane pore-forming active substance that causes cell membrane damage and causes inflammatory factors, such as interleukin-18 (IL-18) and IL-1β are released to cause inflammation. The mechanism of pyroptosis is involved in crystal-related nephropathy, ischemia-reperfusion kidney injury, diabetic nephropathy, and renal fibrosis. This article will review the specific mechanism of pyroptosis and its role in the occurrence and development of a variety of kidney diseases.
