4 Effect of miRNA-1 and miRNA-133 overexpression on myocardial differentiation of inducible pluripotent stem cells
WANG Fangfang BAI Baobao▲ ZENG Di CHU Yi LI Xue
Objective To investigate the effect of miRNA-1 and miRNA-133 overexpression on myocardial differentiation of inducible pluripotent stem cells (iPSC). Methods Embryos from two-week-old American Institute of Cancer Research (ICR) pregnant mice were collected, 0.5 g/L of trypsin was used for digestion of vaccination, mitomycin was used after spreading to three generations, mouse induced pluripotent stem cells (miPSC) was incubated and cultivated at 37℃, miPSC growed on embryonic fibroblasts was digested and passaged. Medium was replaced, miRNA (U6) (10 nmol/L, marked as control group), miRNA-1 (10 nmol/L, marked as miRNA-1 group), miRNA-133 (10 nmol/L, marked as miRNA-133 group), miRNA-1 and miRNA-133 (10 nmol/L respectively, marked as miRNA-1+133 group) were added and transfected for 24 hours, and were washed by PBS, then miPSC differentiation medium was added, and its differentiation was daily observed, cell pulse rate was calculated. RT-PCR was used to detect the expression of miRNA-1, miRNA-133 and cardiac troponin T (cTnT). Results The cTnT was barely expressed in undifferentiated miPSC, compared with 0 day  of differentiation, the expreesion level of cTnT of miRNA-1 group and miRNA-133 group was increased after 5 days of differentiation (P < 0.05), the increase was more obvious after 10 days of differentiation (P < 0.01). After 10 days of differentiation, the expression level of cTnT of miRNA-1+133 group was obviously higher than those of miRNA-1 group and miRNA-133 group (P < 0.01). Compared with control group, the cell pulse rate of miRNA-1 group and miRNA-133 group were not increased (P > 0.05), while the cell pulse rate of miRNA-1+133 group was significantly increased (P < 0.01). Conclusion miRNA-1 or miRNA-133 alone does not promote myocardial differentiation, only after the co-expression of miRNA-1 and miRNA-133, it can significantly improve the directional differentiation efficiency of miPSC to cardiac cells.

LI Yongli1 ZHAO Jing2 LIU Liming1 ZHANG Quanbo1 LIU Li1 WU Bihua1
Objective To investigate the effect of exogenous insulin-like growth factor-1 (IGF-1) on learning and memory ability in rats with global cerebral ischemia and its possible mechanism. Methods A total of 110 Wistar male rats were divided into normal group, sham operation group, global cerebral ischemia model group, model administration group 1 and model administration group 2; there were 10 rats in each of the normal group and sham operation group. There were 30 rats in each of the whole cerebral ischemia model group, the model drug administration group 1 and the model drug administration group 2. All operated groups were embedded micro-injection catheter into the lateral ventricle, and the whole cerebral ischemia model was established on the 6th day after catheterization with modified Pusinelli four-vessel blockade. Rats in the global cerebral ischemia model group, model drug administration group 1, and model drug administration group 2 were injected with 10 μL of normal saline, IGF-1 (0.2 μg/μL) 10 μL, and IGF-1+PPP (after intraperitoneal injection of blocker PPP 20 mg/kg for 30 min, IGF-1 (0.2 μg/μL) 10 μL was injected into the lateral ventricle), once a day for 7 d. In the sham operation group, NS 10 μL was injected into the lateral ventricle of rats. Morris water maze test was used to test the learning and memory ability of rats in each group before and after administration. HE staining was used to observe the apoptosis of hippocampal CA1 neurons. Immunohistochemistry was used to detect the expression of p-Akt and p-mTOR proteins in hippocampus. Results Compared with the global cerebral ischemia model group, the average escape latency of the model administration group 1 was significantly shortened (P < 0.05), the number of straddle platform and the original platform quadrant swimming time increased significantly (P < 0.05); the number of cells in the hippocampal CA1 area increased. The arrangement was uniform and the structure was clear; the expression of p-Akt and p-mTOR protein were higher (P < 0.05). Conclusion The learning and memory ability in rats with global cerebral ischemia was improved by exogenous IGF-1. This may be related to the inhibition of apoptosis in the hippocampal CA1 region and the enhancement of the expression of p-Akt and p-mTOR proteins in the PI3K/Akt signaling pathway.

16 Study on the inhibition effect of Tiaojing Zanyu Pills for oxytocin induced contraction of uterine smooth muscle in vitro in rats
WANG Jingjing1 WANG Zhang2
Objective To observe the effect of Tiaojing Zanyu Pills on isolated uterine smooth muscle in rats. Methods A certain dose of Estradiol Benzoate was administered intraperitoneally to induce allergy in rats. After 24 h, the rats were killed and the smooth muscle of the uterus was removed, and then oxytocin was added to induce constriction in the smooth muscle of the uterus. Finally, suspension with three different concentrations (0.1854, 0.3705, 0.7394 mg/mL) of Tiaojing Zanyu Pills were added respectively. Results Tiaojing Zanyu Pills had no obvious effects on normal rats in terms of the average tension, frequency and activity of uterine smooth muscle contraction (P > 0.05). However, the medium concentration (0.3705 mg/mL) and high concentration (0.7394 mg/mL) of Tiaojing Zanyu Pills could obviously inhibit the oxytocin which thereby induced contraction of uterine smooth muscle in rats, thus resulting in decrease in the average tension and activity (P < 0.05), but no obvious effect on the contraction frequency was observed (P > 0.05). Conclusion Tiaojing Zanyu Pills can directly inhibit the pathological conditions of the uterine smooth muscle contraction, which may be one of the mechanism of clinical treatment of dysmenorrhea.

60 Advances in the interaction of Dax1 with embryonic stem cells multipotential network factors Oct4, Sox2, Nanog, and Esrrb
SUN Huilin YANG Dan HU Zhe▲
The self-renewal and pluripotency of embryonic stem cells (ESCs) are maintained by the large and complex interconnected transcriptional regulatory network formed by transcription factors such as Oct4, Sox2, Nanog, and Esrrb, and Dax1 can also participate in the regulation of ESCs. Dax1 interacts with core factors such as Oct4, Sox2, Nanog and Esrrb, and participates in maintaining characteristics of ESCs and inhibiting ESCs differentiation. Dax1 can also produce a similar promotion to Nanog in the later stages of induction of induced pluripotent stem cells (IPSCs). To study the role, function, status and interaction of Dax1 in the ESCS transcriptional regulatory network and the induction of IPSCs, and the specific mechanisms of interaction with other transcription factors can be helpful for stem cell mechanism research and clinical therapeutic transformation research. At present, there are few studies on the role of Dax1 in embryonic stem cells. This paper reviews the existing research results in order to provide reference for further research and clinical treatment transformation of embryonic stem cells.

Meta-analysis showed that prophylactic use of antimicrobial drugs was associated with the incision infection after episiotomy[OR = 0.50, 95%CI (0.35, 0.71), P = 0.0001]. Subgroup analysis about the modes of action of antimicrobial drugs suggested that the use of focal irrigation had a relevance to the incision infection after episiotomy [OR = 0.25, 95%CI (0.12, 0.53), P = 0.0003], while there was no significant association between systemic application and incision infection [OR = 0.78, 95%CI (0.48, 1.26), P = 0.31]. Conclusion Prophylactic use of antimicrobial drugs may reduce the incidence of incision infections after episiotomy, especially in focal irrigation applications. However, due to the limited number of documents and sample size, large-sample, multi-center and well-designed clinical trials are still needed to confirm and improve the conclusions of this study.
