Establishment of decoction kinetic model of Astragalus membranaceus of Buyang Huanwu Decoction |

Abstract Objective To determine the content of calycosin-7-glucoside in Buyang Huanwu Decoction, and to establish the decoction kinetic model of Astragalus membranaceus of Buyang Huanwu Decoction. Methods The content of calycosin-7-glucoside in Buyang Huanwu Decoction was determined by high performance liquid chromatography. The chromatographic column was Agilent C18 column (250 mm×4.6 mm, 5 μm), mobile phase was acetonitrile (A)-0.05% aqueous solution of phosphoric acid (B) with gradient elution, flow rate was 1 mL/min and detection wavelength was 230 nm, the sample volume was 10 μL. Based on the Fick′s diffusion law, the equation parameters were deduced and fitted. Results Calycosin-7-glucoside was in a good linear relationship in the range of 0.10-0.60 μg, the regression equation was y = 35.95x - 0.3605, R2 = 0.9999, the kinetic equation of decoction was C■=■t■■, the verification result was good. Conclusion The kinetic model established in this study is in good agreement with the experimental results. The standard deviation (0.3013%) of the experimental values and the calculated values under each condition can basically meet the requirements of industrial standard deviation.
