4 Effect of paeoniflorin on vascular endothelial growth factor induced tubule formation of human umbilical vein endothelial cells
CHI Xiaowei1 CHEN Gangling2
Objective To study the effects of paeoniflorin (PF) on vascular endothelial growth factor (VEGF) induced tubule formation of human umbilical vein endothelial cells (HUVECs). Methods The HUVECs cell lines were resuscitated and cultured. MTT assay was used to determine the effects of different concentrations of PF (1, 3, 10, 30, 100 μmol/L) on HUVECs cell viability and the increase of cell viability induced by VEGF. Tubule formation experiments were conducted to evaluate the effect of different concentrations of PF (1, 3, 10, 30, 100 μmol/L) on VEGF induced tubule formation of HUVECs. The effect of PF (1, 3, 10, 30, 100 μmol/L) on VEGF induced HUVECs migration was evaluated by cell scratch test. Results Compared with blank group, there was no statistical significance in the effect of different concentrations of PF (1, 3, 10, 30, 100 μmol/L) on HUVECs cell viability (P > 0.05). Compared with model group (VEGF was given 10 ng/mL, the same as below), there was no statistical significance in the effect of different concentrations of PF (1, 3, 10, 30, 100 μmol/L) on VEGF induced increased HUVECs cell viability (P > 0.05). Compared with model group, different concentrations of PF (1, 3, 10, 30, 100 μmol/L) could inhibit the tubule formation of HUVECs induced by VEGF in vitro. Compared with model group, the scratch width ratio of 24 h/0 h in 10, 30 and 100 μmol/L PF dosage groups were all significantly higher than that in model group, with highly statistical significance (all P < 0.01). Conclusion PF inhibits VEGF induced tubule formation of HUVECs in vitro, and this effect may be realized by inhibiting VEGF induced HUVECs migration.

LI Zaiwang1 CAO Tingting2 WANG Qian1 TU Jingmei1 HAN Jing1
Objective To investigate the effects of epidermal growth factor receptor (EGFR) inhibitor Osimertinib on blood-cord barrier permeability after spinal cord injury (SCI) in rats. Methods A total of sixty eight week-old female SD rats (200 to 250 g) were divided into sham operation group (n = 20) and SCI model group (n = 40) according to random number table method. The 40 SCI model rats were divided into injury group and Osimertinib group by random number table method, with 20 rats in each group. The Osimertinib group was gavaged with 0.4 mg/mL Osimertinib solution (2 mL/d), and the sham operation group and the injury group were gavaged with normal saline (2 mL/d). All the three groups were treated for 3 days. The water content of spinal cord tissue was determined 3 days after SCI, the expression of aquaporin-4 (AQP-4) protein was detected by Western blot, the evans blue (EB) dye leakage test and the albumin content of spinal cord were determined by immunofluorescence technique to evaluate the changes of blood and spinal cord barrier permeability. Basso-Beattie-Bresnahan (BBB) score was used to evaluate the motor function of rats. Results The water content of spinal cord in Osimertinib group at 3 days after SCI was significantly lower than that in injury group (P < 0.05). The expression of AQP-4 protein in Osimertinib group was significantly lower than that in injury group (P < 0.01). The concentration of EB dye in spinal cord tissue in Osimertinib group was less than that in injury group, and the fluorescence intensity of EB was weaker than that in injury group (P < 0.05). The immunofluorescence intensity of albumin in Osimertinib group was weaker than that in injury group. The overall BBB score of Osimertinib group was significantly higher than that of injury group (P < 0.05). Conclusion The EGFR inhibitor Osimertinib can improve the damage of the blood-cord barrier after SCI and alleviate the secondary injury of the spinal cord, and promote the recovery of neurological function.

13 Clinicopathological significance of hMSH2 and FHIT expression in Kazakh esophageal squamous cell carcinoma
Dannier·Duolikun1,2 Kawuli·Jumai3 ZHANG Haiping3 Madiniyet·Niyaz1 Edris·Awut2,3
Objective To investigate the expression of hMSH2 and FHIT genes in Kazakh esophageal squamous cell carcinoma, and analyze the correlation between gene expression and its clinicopathological significance. Methods Esophageal cancer and its adjacent normal tissue specimens were selected from 43 patients who underwent radical resection of esophageal cancer in the First Affiliated Hospital of Xinjiang Medical University from May 2013 to May 2017. qRT-PCR method was used on 30 cases, while immunohistochemistry was conducted on 43 cases to study their clinicopathological characteristics. Results The mRNA expression levels of both hMSH2 and FHIT genes in esophageal squamous cell carcinoma tissues were significantly lower than those in adjacent normal tissues, and the differences were statistically significant (P < 0.05). Results of immunohistochemistry showed that the positive expression rate of hMSH2 and FHIT protein in esophageal squamous cell carcinoma tissues was significantly lower than that in adjacent normal tissues, and the difference was statistically significant (P < 0.05). The expression level of HMSH2 protein was related to the degree of tumor differentiation, lymph node metastasis and postoperative pathological stage (P < 0.05), while it had nothing to do with the age, sex and tumor location of the patients with esophageal cancer (P > 0.05). The expression level of FHIT was only related to the depth of infiltration (P < 0.05). Kaplan-Meier survival analysis found that the expression level of HMSH2 in esophageal cancer was correlated with survival rate (P < 0.05). The results of COX multivariate survival analysis showed that the loss of hMSH2 gene expression and the degree of tumor differentiation were independent risk factors for poor prognosis in Kazakh patients with esophageal squamous cell carcinoma (P < 0.05). Conclusion hMSH2 and FHIT may be involved in the occurrence and development of Kazakh esophageal cancer, so the detection of co-expression could be used as an important indicator for prognosis.

HUANG Jiali CHEN Zhenyao LEI Tianyao WANG Zhaoxia CHENG Zhixiang
Objective To construct human non-small cell lung cancer (NSCLC) Osimertinib-resistant cells HCC827OR in vitro, and to explore its resistance mechanism. Methods Using the NSCLC cell line HCC827, the Osimertinib-resistant cell line HCC827OR was constructed by the concentration-increasing method, and the differences in morphology, proliferation, apoptosis, and cell protein expression between the two cell lines were compared. Results Half-inhibitory concentration (IC50) and resistance index (RI) results showed that HCC827OR was highly resistant to Osimertinib, and the morphology of HCC827OR and HCC827 cells were significantly different. On the first day of the CCK8 proliferation experiment, the cell viability of HCC827OR was higher than that of HCC827 (P < 0.05), and on the second, third and fourth days, the cell viability of HCC827OR was significantly higher than that of HCC827 (P < 0.01). In the clone formation experiment result showed that, the colony formation number of HCC827OR was higher than that of HCC827 (P < 0.01). After EdU staining, the number of EdU-positive cells in HCC827OR was higher than that in HCC827 (P < 0.01). In the apoptosis experiment result showed that, compared with HCC827, the apoptosis of HCC827OR cells was reduced (P < 0.01). In Western blot experiment, compared with HCC827, the expression of EGFR and p-EGFR in HCC827OR cells decreased, and the expression of p-AKT, p-ERK and Vimentin increased. Conclusion The NSCLC osimertinib-resistant cell line HCC827OR is successfully constructed. It is speculated that the resistance mechanism is related to the decreased expression of EGFR and p-EGFR, and the increased expression of p-AKT, p-ERK and Vimentin.

XU Jingxuan1,2 FU Qiuju3 LI Liang1 QIAN Xiaoping4
Objective To analyze the research situation of traditional Chinese medicine in the treatment of radiation enteritis in recent 10 years by visualization method. Methods Articles related to the treatment of radiation enteritis with traditional Chinese medicine were collected in China national knowledge infrastructure (CNKI) from January 2010 to January 2020. CiteSpace was used to perform co-occurrence analysis and visualize the collaboration networks of the authors, institutes, keywords, in order to understand the research status, hotspots and trends in this field. Results A total of 193 articles were included. The annual number of published articles on the whole showed a trend of increasing first and then decreasing slightly. The author who published the most articles was Wang Yuguo, among whom Li Mei’s team and Wang Lei’s team worked closely together. Seven institutions were the main bases for the study of traditional Chinese medicine treatment of radioactive enteritis. Research institutions were independent and scattered, and no obvious cooperative network had been formed. The keyword co-occurrence network formed five clusters. When the keywords related to the subject words were removed, the top two keywords in frequency were “retention enema” and  “Chinese medicine enema” respectively. The top three keywords with high mediating centrality were “cervical cancer” “retention enema” and “Chinese medicine enema”. Conclusion In the past ten years, the number of related studies on traditional Chinese medicine treatment of radiation enteritis with is not large, with relatively few researchers involved, scattered research institutions and lack of cooperation. There are few studies on the mechanism of action and the research depth is not enough. Therefore, high-quality studies should be carried out to provide evidence support for clinical practice. The clinical effect of traditional Chinese medicine retention enema on radiation enteritis is the focus of research.

32 New progress of sodium-glucose cotransporter 2 inhibitors in the treatment of diabetic nephropathy
REN Yanrong1,2 YU Lei2▲
Diabetic nephropathy (DN) is a common and serious microvascular complication of diabetes mellitus, and it is the leading cause of end-stage renal disease (ESRD). Despite the use of various drugs such as renin-angiotensin system inhibitors, DN often continues to progress, and patients are at great risk of progressing to ESRD. Sodium-glucose cotransporter 2 inhibitors (SGLT-2i) are a novel class of target drugs that have been extensively investigated in type 2 diabetes mellitus (T2DM). The inhibitors block sodium and glucose reabsorption in the renal proximal tubule, leading to glycosuria for the treatment of hyperglycemia. Recent large scale clinical trials in T2DM patients have shown that SGLT-2i exhibit cardiorenal protective propertiesin in addition to its hypoglycemic effects. Especially in the patients complicated with DN, SGLT-2i are found to protect renal function and reduce urinary albumin excretion through various mechanisms. Therefore, the latest studies have discovered that SGLT-2i can significantly improve the prognosis of patients with DN. This article reviews the recent progress on SGLT-2i for renal protective effects and mechanisms in DN.

36 Research progress on the relationship between estrogen receptor-α36 and breast cancer
HOU Mengyue HUANG Zhaoquan
Estrogen receptor (ER) -α36 is a 36 kD variant of ER-α66, a subtype of ER-α. ER-α36 is mainly expressed in cytoplasm and cell membrane. Compared with ER-α66, it lacks the transcriptional activation domain (AF) -1 and AF-2, and retains the DNA binding domain, partial dimerization domain, and partial distributive binding domain of ER-α66. It can cooperate with various signaling factors such as protein kinase A, protein kinase C and Ca2+, and can rapidly activate non-genomic estrogen signaling pathway in a short period of time to regulate the biological behavior of tumors. Current studies have found that ER-α36 is expressed in human breast cancer, endometrial cancer and other estrogen-related tumor cells. Therefore, the relationship between ER-α36 and malignant tumor is worth studying.

39 Progress in the treatment of dyskinesia in Parkinson’s disease
ZHONG Zhe XU Min CHEN Weijie GAO Huan YE Min
Levodopa-induced dyskinesia is a complication caused by long-term treatment of Levodopa in patients with Parkinson’s disease. The treatment of Levodopa-induced dyskinesia is mainly divided into two parts, drug therapy and non-drug therapy. In non-drug therapy, deep brain stimulation and unilateral pallidotomy is considered to be effective. Drug therapy focuses on providing continuous dopaminergic stimulation, such as the development of new Dopamine preparations, catechol-O-methyltransferase inhibitors, Subcutaneous Apomorphine and Levodopa-carbidopa enteral gel continuous jejunal infusion, the second is the use of non-dopaminergic drugs such as Amantadine, Clozapine and Levetiracetam. Although the preclinical results of most non-dopaminergic drugs are encouraging, however, due to the uncertainty of its effect and its side effects, at present, the transformation to clinical practice is still challenging. This article describes in detail the latest treatment of Levodopa-induced dyskinesia at home and abroad, so as to provide reference for daily clinical work.

Objective To analyze the risk factors of pulmonary thromboembolism with different severity after craniocerebral tumor operation under cluster intervention measures, and to provide reference for early clinical intervention of pulmonary thromboembolism. Methods The clinical information of 56 patients with pulmonary thromboembolism diagnosed by CT pulmonary angiography after craniocerebral tumor operation in the Department of Neurosurgery, Sanbo Brain Hospital, Capital Medical University from June 2016 to June 2019 was retrospectively analyzed. The patients were divided into low risk group (36 cases) and high risk group (20 cases) according to the diagnosis and treatment guidelines of pulmonary thromboembolism. Clinical phenotypes, coagulation function and perioperative related factors were compared between the two groups, and multivariate logistic regression was used to analyze the risk factors of pulmonary thromboembolism with different severity. Results Among the 56 patients, 35 were male and 21 were female. The mean age was (46.23±18.12) years old. Postoperative pathology showed malignant tumor in 20 cases, accounting for 35.71%; benign tumors in 36 cases, accounting for 64.29%. Tumor site classification showed sellar region tumors in 26 cases, accounting for 46.43%; supratentorial tumor in 21 cases, accounting for 37.50%; skull base tumor in 7 cases, accounting for 12.5%; infratentorial tumor in 2 cases, accounting for 3.57%. There were no significant differences in clinical phenotype and coagulation function between the two groups (P > 0.05). There was no significant difference in perioperative related factors between the two groups except the time of water fasting before operation (P > 0.05). Multivariate logistic regression analysis showed that the presence of postoperative limb mobility disorder was a risk factor for pulmonary thromboembolism after craniocerebral tumor operation (OR = 9.574, P = 0.02). Conclusion Pulmonary thromboembolism after craniocerebral tumor operation has complex underlying causes and no specific clinical manifestations. The identification of the risk factors of pulmonary thromboembolism combined with the results of laboratory examination is helpful for the early diagnosis and treatment of  pulmonary thromboembolism.

81 Analysis of influencing factors of the formation of postoperative acute pressure ulcers in patients with subarachnoid hemorrhage
SHEN Jie1 WANG Jing2▲
Objective To investigate the influencing factors of the formation of postoperative acute pressure ulcers in patients with subarachnoid hemorrhage (SAH). Methods The data of 80 patients with SAH who completed surgery in Xinhua Hospital Affiliated to Shanghai Jiao Tong University School of Medicine from January to October 2020 were retrospective analyzed. According to whether postoperative acute pressure ulcers occurred, they were divided into acute pressure ulcers group (10 cases) and no acute pressure ulcers group (70 cases). The general data of the two groups were compared, and the influencing factors of the formation of postoperative acute pressure ulcers in SAH patients were analyzed by univariate and multivariate analysis. Results The intraoperative body temperature of the acute pressure ulcers group was lower than that of the no acute pressure ulcers group, the preoperative fasting blood glucose level was higher than that of no acute pressure ulcers group, and the stress response degree was worse than that of the no acute pressure ulcers group, with statistical significant differences (all P < 0.05). The results of univariate and multivariate logistic regression analysis showed that high intraoperative body temperature were the protective factors for the formation of postoperative acute pressure ulcers in SAH patients (OR < 1, P < 0.05); high level of preoperative fasting blood glucose and severe stress response degree might be risk factors for postoperative acute pressure ulcers in SAH patients (OR > 1, P < 0.05). Conclusion Some patients with SAH are still at risk of postoperative acute pressure ulcers, which may be related to the high level of preoperative fasting blood glucose and severe stress response degree, and the high intraoperative body temperature may be helpful to prevent the formation of postoperative pressure ulcers. Patients at high risk should be actively intervened in early stage.

189 Question and resolution on the qualification management system of Chinese physicians in double track system
ZHANG Yuan1,2 BAI Simin1,2 OUYANG Jing1,2 LI Xiuqin1,2 YUAN Ruihua3
Law of the People’s Republic of China on Traditional Chinese Medicine and Pharmacy advocates the qualification examination system of traditional Chinese medicine doctors, and breaks the single qualification examination system of Law of the People’s Republic of China on Medical Practitioners. At present, the two kinds of physician qualification management systems are both legal and effective, which are separated and parallel, forming a operation mode of double track system. By comparing the two systems, it is found that they are quite different. In operation, it is easy to cause disputes of fairness and risk, and the scope of practice of traditional Chinese medicine practitioners is not clearly stipulated by the two systems. The implementation of the new policy will bring more staff and clinics, which will also bring pressure to grass-roots supervision. In addition, the different interpretation and implementation of the new policy will not be conducive to the unity of legal construction and policy implementation. This paper proposes and approves the following four suggestions: It is suggested that the access system should be strictly enforced to ensure fairness. It is suggested to perfect the regulation of the scope of practice of traditional Chinese medicine practitioners. It is suggested to perfect the management organization and establishment of traditional Chinese medicine practitioners, as well as to perfect the policies with specific expertise, so as to promote the unity of legal construction and policy implementation, to promote the improvement of the management system of traditional Chinese medicine practitioners’ qualification.
