DOI: 10.20047/j.issn1673-7210.2023.09.01
Objective To analyze and identify the interaction between ten-eleven translocation protein 3 (TET3) and O-linked-β-N-acetylglucosamine transferase (OGT) in the nuclei of rat spinal cord neurons. Methods The spinal cord neurons of rats were primarily cultured and total RNA was extracted, and TET3 and OGT gene fragments were obtained by RT-PCR. The tag proteins of Flag and HA were fused to the carboxyl end of TET3 and OGT protein coding sequences by DNA recombination technology. The recombinant fusion protein plasmids of pcDNA3.0-TET3-Flag-HA and pcDNA3.0-OGT-Flag-HA were verified by double enzyme digestion, and then transfected into the rat spinal cord neurons. The nucleoproteins of spinal cord neurons were extracted 48 hours later. The fusion proteins of TET3-Flag-HA and OGT-Flag-HA were verified by Western blot; the protein complexes bound to TET3 and OGT were enriched by Tandem affinity purification, the candidate proteins interacting with TET3 and OGT were screened by electrophoresis and mass spectrometry, and verified by co-immunoprecipitation. Results It was confirmed that the recombinant fusion protein plasmids of pcDNA3.0-TET3-Flag-HA and pcDNA3.0-OGT-Flag-HA were successfully constructed, and the fusion proteins of TET3-Flag-HA and OGT-Flag-HA were expressed in the nuclei of rat spinal cord neurons. In the nuclei of rat spinal cord neurons, the protein interacting with TET3 was OGT, and the proteins interacting with OGT were TET3 and retinoblastoma binding protein 5. Conclusion TET3 and OGT form a complex in the nuclei of rat spinal cord neurons, which may be involved in regulating the transcriptional and expression of some functional proteins.

SUN Yibin1 ZHU Mingxing2 LIU Yueyang3 ZHAO Ruowei1 HUANG Mingqing1 CHENG Zaixing1 ZHENG Yanfang1 LIN Yanxiang1
DOI: 10.20047/j.issn1673-7210.2023.09.07
The incidence of liver injury is increasing year by year, it is urgent to develop high quality hepatoprotective drugs. Traditional Chinese medicine has unique advantages and potential in liver protection, and has therapeutic effect in a variety of liver injury caused by chemical, drug, alcohol, and immunity reasons. Traditional Chinese medicine can alleviate liver injury by improving liver antioxidant capacity, inhibiting hepatocyte apoptosis, regulating hepatocyte autophagy, regulating liver metabolism, and improving liver inflammation. It acts on Nrf2, PI3K, TGF-β1, NF-κB and other targets, and related signaling pathways. Some drugs or active ingredients can act through multiple pathways and multiple targets. Traditional Chinese medicine has a good application prospect in liver protection, but there are many components of the compound and unclear mechanism interaction. Further research is needed to provide reference for clinical application.

Current situation and prospect of exosomes application in tumor diagnosis and treatment
ZHU Xiaoxian1 CHEN Huizhi1 CUI Kuiyan1 ZHONG Wen2 PENG Xinsheng1 ZHOU Yubin1
DOI: 10.20047/j.issn1673-7210.2023.09.08
Exosomes are membranous vesicles with a size of 30-150 nm that are produced by the endovascular process of the endocytic membrane. Exosomes contain a large number of biological macromolecules, such as various enzymes, proteins, mRNA, and non-coding RNA, which make them increasingly regarded as important carriers in intercellular communication and substance exchange. Exosomes are closely related to the processes of tumor growth, angiogenesis, and tumor immune escape, and metastasis, and the research on exosomes in the field of tumors is one of the current hotspots. Because exosomes contain numerous information of donor cells and carry a variety of important biomolecules, they can be applied as biomarkers, drug carriers, or immunomodulators for tumor diagnosis and treatment. In this review, the basic concept, formation and elemental composition, isolation and characterization of exosomes, as well as their applications in tumor diagnosis (as biomarkers) and treatment (as drug carriers and as immunomodulatory substances) are reviewed, and their prospects are discussed.

Research progress of immune regulation mechanism of traditional Chinese medicine on spinal tuberculosis based on the theory of “killing insects and replenishing deficiency”
LIANG Jian1 KANG Wenting1 FENG Jing2 PU Feifei2 XIA Ping1
DOI: 10.20047/j.issn1673-7210.2023.09.09
In traditional Chinese medicine, tuberculosis is mainly caused by “the deficiency of the body and the external sensation of tuberculosis”, and “killing insects and replenishing deficiency” is the general treatment principle of traditional Chinese medicine in the treatment of tuberculosis. Spinal tuberculosis is mostly secondary to pulmonary tuberculosis and digestive tract tuberculosis. Modern medical studies have shown that the pathogenesis and outcome of spinal tuberculosis are closely related to the immune status of the host and the growth and inhibition of Mycobacterium tuberculosis. In the treatment of spinal tuberculosis, single traditional Chinese medicine components, extracts, and traditional Chinese medicine compounds can play an important auxiliary role by regulating the immune function of the body, destroying and inhibiting the generation of pathogenic bacteria, regulating autophagy and apoptosis of cells, and other different immune axes. Based on the theory of “killing insects and replenishing deficiency”, this paper discusses the research progress of the immune regulation mechanism of traditional Chinese medicine on spinal tuberculosis.

DOI: 10.20047/j.issn1673-7210.2023.09.24
Objective To mine the data of ancient and modern medical cases of apoplexy by using statistical analysis and social network analysis, and to analyze the law of their identification and treatment of apoplexy. Methods The medical case data were retrieved, collected, and organized from the super star electronic book database, and the retrieval period was from inception to December 2022, while the medical case data were screened and extracted in conjunction with the National Joint Catalogue of Chinese Medicine Books, and statistical analysis and data mining were performed on the high incidence syndrome type, nature and flavour of traditional Chinese medicine, channel tropism of apoplexy and so on. Results A total of 404 medical cases of apoplexy were included, involving 48 medical case collections, 340 Chinese herbal medicines, 4 937 total medication frequencies, three high frequency symptom types (syndrome of blood stasis due to qi deficiency, syndrome of wind-phlegm obstructing collateral, syndrome of stirring wind due to yin deficiency). The drug natures were usually warm, plain, and cold. The drug flavours were mainly sweet, bitter, and pungent. The drug channel tropisms were liver, spleen, and heart channels. There were ten high centrality value drugs. The results of the social network analysis showed that there was a strong association between the type of symptoms and the formulas and herbal medicines. The treatment was mainly composed of Buyang Huanwu Decoction, Dihuang Yinzi, and other prescriptions. Conclusion Ancient and modern medical cases suggest that apoplexy is closely related to the liver, spleen, and heart. Syndrome of blood stasis due to qi deficiency, syndrome of wind-phlegm obstructing collateral, syndrome of stirring wind due to yin deficiency are the core syndrome types. There is an obvious trend of concentration in the use of drugs, such as invigorating qi, promoting blood circulation for removing blood stasis, and suppressing hyperactive liver for calming endogenous wind, which can provide certain reference for clinical prescription drug use.

Professor Chen Ruixue’s experience in treating thin endometrium infertility based on the theory of “deficiency of qi and stagnation”
GUO Li1 SUI Juan2 REN Mengxue2 ZHOU Lin2 CHEN Ziwei1 BI Hong2 CHEN Ruixue2
DOI: 10.20047/j.issn1673-7210.2023.09.26
This article summarizes Professor Chen Ruixue’s experience in treating thin endometrium infertility based on the theory of “deficiency of qi and stagnation”. Professor Chen believes that “deficiency of qi” and “stagnation” are the main causes of thin endometrial infertility. Among them, “deficiency of qi” is mainly responsible for kidney deficiency and spleen deficiency, and “stagnation” includes qi stagnation, phlegm obstruction, and collaterals stasis. In terms of treatment, Professor Chen advocates that tonifying the kidney and strengthening the spleen to regulate the deficiency qi, soothing liver qi, removing phlegm and dampness, and dissipating blood stasis to smooth the stagnation, especially emphasizes that tonifying kidney and raising the Yuan and promoting blood circulation for removing obstruction in collaterals are the core treatment principles of this disease. Clinically, Professor Chen uses Bushen Yangxue Zhuyun Prescription add or subtract medicine, stages medicine according to the menstrual cycle, combines internal and external application, and the efficacy is outstanding.

Professor Zhou Hua’s experience in treating atrial fibrillation based on staged dialectical theory
TANG Qiaolin QU Huiyan LIU Wenrui YANG Tianshu YANG Tao ZHOU Hua
DOI: 10.20047/j.issn1673-7210.2023.09.27
Atrial fibrillation is one of the most common arrhythmias characterized with high morbidity, disability, and mortality in the clinical field. Professor Zhou Hua believes that atrial fibrillation is the syndrome of deficiency of root case and excess of manifestation. The deficiency of root case is mainly caused by deficiency of qi, blood, yin and yang, the excess of manifestation is mainly phlegm and blood stasis. In terms of treatment, it is emphasized that treatment should be carried out by three stages, including early stage, middle stage, and late stage. At the early stage, the heart’s qi and yin injuries are the key to its pathogenesis, and the treatment is mainly to replenish qi and nourish yin. In the middle stage, phlegm and blood stasis are caused by deficiency of heart and spleen, which should be treated by resolving phlegm, removing blood stasis, and strengthening the spleen. In the later stage, the deficiency of heart and kidney yang caused by the injury of kidney qi is an important reason for the disease to persist and worsen, treat with warming yang and tonifying kidney.

Summary of Professor Cai Lianxiang’s experience in treating unruptured ectopic pregnancy
GAO Shanfeng CHEN Yanxia HUANG Yuxiao▲
DOI: 10.20047/j.issn1673-7210.2023.09.30
Ectopic pregnancy is one of the common acute abdominal diseases in obstetrics and gynecology. Clinically, it is often divided into unruptured type and ruptured type, and the unruptured stage is the best time for traditional Chinese medicine intervention. Professor Cai Lianxiang believes that the pathogenesis of unruptured ectopic pregnancy in traditional Chinese medicine always belongs to the deficiency of root cause and the excess of manifestation, and intermingled deficiency and excess, in which the deficiency of qi and blood is the root cause and the blood stasis and heat is the manifestation. In terms of treatment, the unruptured ectopic pregnancy was treated by stages. In the first stage, the treatment was mainly to promote blood circulation and kill embryos; in the later stage, the treatment was mainly to eliminate symptoms and resolving hard mass. At the same time, in the later stage of treatment, on the basis of internal decoction, external application or enema of traditional Chinese medicine for removing stasis and clearing collaterals was often used to promote lump absorption, prevent pelvic adhesion, and promote fallopian tube recovery patency. Professor Cai is good at combining modern pharmacological research and drug use to provide psychological counseling for patients with ectopic pregnancy, and the idea of strengthening vital qi to eliminate pathogenic factor and protecting and maintaining vital qi throughout the treatment, so as to restore and retain the fertility function of patients to the greatest extent.

Comments on the Abridged English Versions of Qian Jin Yao Fang
YANG Guangyi1 WANG Shanshan2 ZHANG Sijie2 BAI Bing2
DOI: 10.20047/j.issn1673-7210.2023.09.31
Qian Jin Yao Fang, one of the classics of Sun Simiao in Tang Dynasty, has high academic value. Till now, only three abridged English versions have been officially published, which are translated by scholar Ms. Sabine Wilms at Arizona State University in the United States. This paper analyzes the three versions, finding they are characterized by reasonable compilation, various translation, and detailed annotation. As to translation strategies, both “domestication” and “foreignization” are taken into account with specific translation methods such as transliteration, literal translation, and liberal translation, etc. Annotation methods like in-text citations, footnotes, and endnotes are also included. Despite occasional mistranslations and other considerable issues, these versions still retain important academic value. Through the analysis of translation ideas, strategies and methods of the three versions, this paper aims to offer references for the subsequent complete translation work on Qian Jin Yao Fang and the overseas dissemination of traditional Chinese medicine classics.
