ZHANG Yuchun1,2 ZHANG Qing1 YANG Yu1 ZHAO Yijing1 WANG Kun1 MA Xianghua2
Objective To research the influence of Doxorubicin loaded melanin nanoparticles (DOX-MNPs) on drug resistance of anaplastic thyroid cancer (ATC) cells. Methods Dopamine-melanin nanoparticles (MNPs) were synthesized as previously reported and Doxorubicin (DOX) was loaded onto MNPs. The loading efficiency and capacity of MNPs at different mass ratios of DOX to MNPs were determined by UV-Vis spectroscopy and the characterization of nanoparticles was acquired by transmission electron microscopy (TEM) and Brookhaven Zeta PALS analyzer. HTh74 and HTh74R cells were incubated with various concentrations of free DOX and DOX-MNPs (0, 10, 20, 40, 80, 160 mg/L). Cell viability was determined by MTT, and intracellular DOX fluorescent signal was determined by flow cytometry studies. Results The MNPs was successfully synthesized and the loading efficiency of DOX-MNPs decreased with the increasing mass ratio of DOX to MNPs. The loading efficacy peaked at 93.45% when the mass ratio was 0.167:1. At this mass ratio, the obtained DOX-MNPs had similar morphology to MNPs, and zeta potential and size distribution of DOX-MNPs were 13.79 mV and (241.1±4.8) nm. In HTh74R cells, when the concentration of DOX-MNPs was higher than 20 mg/L, the cell viability was significantly lower than that of the equivalent drug concentration (P < 0.05), and the uptake rate of DOX-MNPs was significantly higher than that of free DOX (P < 0.05). The degree of uptake was similar to that of HTh74 cells. Conclusion The cellular uptake capacity and therapeutic effect of the DOX-MNPs are significantly higher than those of free DOX at the same drug concentration in drug-resistant HTh74R cells.

LIU Lei1 LI Yan2 GUO Yuanbiao1
Objective To investigate the difference of serum SALL4 levels between gastric cancer patients and healthy people, and to explore the role of SALL4 in the progression of gastric cancer. Methods A total of 47 patients with gastric cancer who were treated in the Third People′s Hospital of Chengdu from March 2016 to April 2018 and 29 healthy volunteers who participated in the physical examination were selected. The serum levels of SALL4 were detected by ELISA. The expression of SALL4 in mRNA level and protein level in various gastric cancer cell lines and gastric mucosa cells were analyzed by Real-time PCR and Western blot respectively. The effects of SALL4 on the cell migration and invasion ability were analyzed by siRNA interference. The expression levels of related molecules of epithelial-mesenchymal transition (EMT) were detected by Western blot. Results The serum SALL4 levels in patients with gastric cancer were significantly higher than those in healthy volunteers, and the difference was highly statistically significant (P < 0.0001). The expression of SALL4 mRNA and protein in various gastric cancer cell lines was higher than that in gastric mucosal cell lines. The migration and invasion ability of gastric cancer cells were significantly decreased after interfering SALL4 expression, and the differences were statistically significant (P < 0.05). Decreased expression of SALL4 leaded to the up-regulation of E-cadherin expression and the down-regulation of snail, β-catenin and Wnt3a. Conclusion SALL4 is related to the progression of gastric cancer. It may promote cell migration and invasion by inducing the EMT process in gastric cancer cells.

29 Research progress of the mechanism of drug resistance and detection methods of Mycobacterium tuberculosis
SONG Jing CHE Nanying
This article reviews the mechanism and detection methods of drug resistance in Mycobacterium tuberculosis(MTB). The mechanism of drug resistance is divided into intrinsic resistance mechanism of cell wall permeability reduction and efflux pump action and acquired resistance mechanism of target gene mutation. With the development of molecular biology techniques, mutations in target genes are considered to be the main mechanism of drug resistance for MTB. The main detection methods for drug resistance are phenotypic susceptibility testing and molecular susceptibility testing. The phenotypic assay method is used as the golden standard, but the detection period is long, while gene mutation detection can be completed within a few hours, so there is a better application prospect in the diagnosis of tuberculosis. In this paper, the mechanism and detection methods of drug resistance in MTB are reviewed in order to provide some reference for the development of rapid molecular diagnostic tools and anti-tuberculosis drugs.

SUN Yanling ZHANG Wei GENG Hui
Based on the particularities of community traditional Chinese medicine (TCM) health elderly care service, to explore the O2O platform model of Yunnan Province community TCM health care service, which combines the communication of TCM health care culture with the promotion of TCM health literacy and TCM health assessment with TCM health services. However, the implementation of O2O platform is restricted by a series of factors, such as low informations level of grass-roots community health service institutions, urgent need to establish multi-party cooperation mechanism, and insufficient TCM health literacy of elderly people in the community. At the present stage, the key to promot the development of the O2O platform is to strengthen the concept and skills of TCM health care for the elderly in the community. At the same time, the government should play the role of guidance and supervision, promote the integration of the TCM health care service industry in the community, and improve the TCM health service capacity in the community.
