WU Jing1,2 WANG Jingjing2 LI Xiaojie2 HE Jie2,3
Objective To investigate the expression and clinical significance of the oncogene IMP3 and sialomucin CD164 in glioblastoma (GBM) tissues. Methods One hundred and nineteen GBM paraffin-embedded samples from January 2011 to July 2015 in Anhui Provincial Hospital were collected. The expression of IMP3 and CD164 was detected by automated immunohistochemistry. The results were analyzed with clinical and pathological factors and the survival of patients. Results The high expression rates of IMP3 and CD164 were 69.7% (83/119) and 81.5% (97/119) respectively. There was a positive correlation between the two expression levels (r = 0.252, P = 0.006). In the connected analysis with clinical data, the expression of IMP3 was positively correlated with EGFR state (r = 0.242, P = 0.008), and was not significantly related to the gender, age, tumor size, tumor location and Ki67 proliferation index (P > 0.05). CD164 expression was positively correlated with age (r = 0.285,P = 0.002) and EGFR (r = 0.249,P = 0.006), not other factors (P > 0.05). The overall survival (OS) of patients with CD164 high expression were significantly shorter than those with low expression (P = 0.004). There was no significant correlation between IMP3 and OS alone (P = 0.056). Age and CD164 were independent prognostic factors of GBM (P = 0.036, 0.005). Conclusion The expression of CD164 in GBM is closely related to IMP3, and CD164 can be used as an independent predictor of poor prognosis of GBM.

Objective To explore the effects of chemotherapeutic drugs non-myeloablative pretreatment and human cord blood hematopoietic stem cells transplantation (HSCT) on the spermatogenic function in mice. Methods Thirty Balb/c mice were divided into 3 groups by random digital table method, with 10 mice in each group, which contained control group, Busulfan (BU) combined with Cyclophosphamide (CY) non-myeloablative conditioning (BU+CY) group and HSCT group. In addition to the control group, BU+CY group and HSCT group were pretreated with BU 10 mg/kg and CY 120 mg/kg to induce testicular lesion, and HSCT group was taken human umbilical cord blood HSCT. After HSCT for three weeks, the testicular index was calculated by weighting method. The routine quality analysis of semen and histopathological examination of the testis and epididymis were carried out. Results Compared with the control group, the testicular index, sperm density and sperm viability in BU+CY group were significantly decreased, but the sperm malformation rate was significantly increased (P < 0.01). The graft versus host disease (GVHD) score was higher in HSCT group. The testicular index, sperm density and sperm viability in HSCT group were higher than those in BU+CY group (P < 0.05), while the sperm malformation rate was lower than that of BU+CY group (P < 0.01). The pathological examination found that the testicular volume in BU+CY group was smaller, the seminiferous tubules were obviously impaired, the spermatogenic cells were swelled and arranged in disorder, the seminiferous epithelia was thinned, the number of sperm was significantly reduced in seminiferous tubule and epididymal tubule. Compared with the BU+CY group, the testicular volume in HSCT group was increased, the seminiferous tubule was more integrity, and the number of sperm was increased, but the number of layers seminiferous epithelia cells and spermatocyte were still less. Conclusion BU+CY non-myeloablative conditioning can cause obvious testis injury. Cord blood HSCT can promote a certain extent recovery of testicular spermatogenesis in mild acute GVDH mice.

ZHOU Min HU Ming HE Songming HUANG Guotao CHEN Qian HONG Li
Objective To explore a culture of SCs method with higher cell purity and shorter time by comparing the two methods including the double-enzyme digestion and single-enzyme digestion of primary culture of Schwann cells (SCs) using the neonatal rats. Methods Bilaterally sciatic nerves were dissected from 4 to 5-day-old neonatal SD rats and double-enzyme digestion method and single-enzyme digestion method which both combined with hemi-explants-culture method, differential digestion method, differential adhesion method, and cytosine arabinoside were used to culture and purify SCs, respectively. The morphology of the SCs was observed under the inverted microscope at 48 hours and 6 days of culture. Then the SCs were digested and passaged after 7 days in culture and counted under the cell counter. The purity of SCs was identified by S-100 immunofluorescence staining. CCK-8 was used to measure the P3 cells growth curve, respectively. Results With the same number of neonatal rats, double-enzyme digestion method acquired SCs at least twice than single-enzyme digestion method. (95.29±1.71)% of S-100 positive SCs cultured were shown by immunofluorescence staining in the double-enzyme digestion method group, and (72.41±2.16)% in the single-enzyme digestion method group (P < 0.05). There was no difference between the two cultures in the growth curve of P3 cells (P > 0.05). Conclusion The double-enzyme digestion combined with hemi-explants-culture method, differential digestion method, differential adhesion method, and cytosine arabinoside may obtain sufficient amount of high-purity SCs in a short time and be applied in the further cytological experimental research on peripheral nerve regeneration.

LIU Na1 QI Yanhua1 ZHOU Qi1 CHENG Chuantao2 YANG Wenbin2
Objective To explore the expression of long non-coding RNA H19 (lncRNA H19) in thyroid carcinoma (TC), and its correlation with the clinicopathologic characteristics and prognosis of TC. Methods From January 2009 to January 2012, 131 TC tissues and adjacent tissues were collected from patients after thyroid surgery between as case group. Concurrently, additional 122 normal thyroid tissues from patients with benign thyroid lesions were obtained as normal control group. The expression level of lncRNA H19 were detected by quantitative real-time PCR (qRT-PCR). five years follow-ups were conducted. Receiver operating curve(ROC) was used to evaluate the diagnostic values of lncRNA H19 for TC. The Kaplan-Meier method were employed to analyze 5-year survival rate of TC patient. Both univariate and multivariate analyses were performed to analyze the prognostic factors of TC. Results The lncRNA H19 expression was higher in TC tissues than that in the adjacent and normal tissues(P < 0.05). The area under the ROC curve(AUC) of lncRNA H19 were 0.801. Diagnostic boundary point was 3.58. Sensitivity and specificity was 72.5% and 75.4% respectively. Youden index was 0.479. The lncRNA H19 expression in TC tissue with tumor diameter no less than 1 cm, TNM Ⅲ+Ⅳ stage, or lymph node metastasis was evidently higher than that in TC tissue with tumor smaller than 1 cm, TNM Ⅰ+Ⅱ stage, or without lymph node metastasis(P < 0.05). The 5-year survival rate of TC patients with high lncRNA H19 expression declined (28.44%). The expression of lncRNA H19, tumor diameter, TNM stage and lymph node metastasis were independent prognostic factors of TC patients (P < 0.05). Conclusion The increased lncRNA H19 expression levels are associated with clinicopathologic characteristics and poor prognosis in TC patients. And it may be one of the molecular regulation mechanism in TC occurrence and development. This study also can provide basis for noninvasive diagnosis and therapeutic target searching in TC.

Advanced in the research of miR-375 on tumor development and diagnosis
SUI Chengliang1 HUANG Zhaoquan2
MicroRNAs (miRNAs) are conserved small non-coding RNAs that play important roles in various biological processes via controlling gene expression. Among them, miR-375 is firstly identified in pancreatic β-cells and proved to regulate insulin secretion and islet formation. Furthermore, it turns that miR-375 is involved in cancer development and progression by targeting several important oncogenes including AEG-1, YAP1, IGF1R and PDK1. Unlike some other cancers, the levels of miR-375 in prostate and breast cancer specifically increase, indicating its action in tumor development. Therefore, miR-375 may be a potential target for suppressing tumor growth, and has the clinical value as a biomarker for early diagnosis and prognosis prediction.

HU Zhengrong1 WEI Binhua1 LI Changke1 TAN Jiangbo2 WU Qinru3
Objective To investigate the effect of low central venous pressure (LCVP) combined with acute hypervolemic hemodilution (AHHD) on intrapulmonary shunt and arterial oxygen partial pressure during one lung ventilation in thoracic surgery. Methods From January 2015 to December 2017, ASA grade Ⅰ-Ⅱ undergoing elective radical resection of lung cancer and 50 thoracic surgery patients in the Affiliated Yuebei People's Hospital, Shantou University Medical College, were randomly divided into two groups: group AHHD with LCVP (group LA) and control group (group C), each group had 25 cases. Group LA was given the limited crystalloid the amount of nitroglycerin and continuousmicro pump injection method after anesthesia in advance of LCVP technology, control of CVP in 2-5 cmH2O, then combined with the application of AHHD technology, rapid infusion of 6% hydroxyethyl starch (130/0.4) 15 mL/kg on blood thinners, keep Hct at 25%-30%. Group C was maintained at the normal level of 6-12 cmH2O CVP. On the each time as 10 min after double lung ventilation in the supine position (T0), 10 min after one lung ventilation in supine position (T1), 10 min after single lung ventilation in lateral position (T2), 10 min after pulmonary lobectomy (T3), the blood glucose and cortisol level were measured by collecting, and the blood of the elbow. The radial artery blood and right atrial mixed venous blood was collected for blood gas analysis, and record blood glucose concentration, serum cortisol level, SpO2, HR, MAP, CVP, BIS, PetCO2, arterial oxygen content (CaO2), mixed venous oxygen content (CvO2), arterial and mixed venous blood oxygen partial pressure (PO2), arterial and mixed venous blood oxygen saturation of hemoglobin (SO2), arterial and mixed venous blood hemoglobin (Hb) etc.. The rate of intrapulmonary shunt (Qs/Qt), Qs/Qt = (CcO2-CaO2)/(CcO2-CvO2)×100% were calculated according to the standard three compartment model of pulmonary blood flow distribution. Results The blood glucose concentration, serum cortisol level, SpO2, HR, MAP, BIS, PetCO2 of patients in two groups at each time point was basically stable, but after one lung ventilation, LA group and group C significantly increased intrapulmonary shunt (P < 0.05), which increased by 13.7% and 14.9% respectively in supine position, lateral position increased 11.4% and 14.3% respectively. arterial oxygen partial pressure of two groups were significantly decreased (P < 0.05), and compared with the supine lateral position decreased more obviously, but there was no significant difference between the two groups (P > 0.05). After lobectomy, the intrapulmonary shunt in group LA and group C was significantly reduced (P < 0.05), 7.2% and 8.6% respectively, and the arterial oxygen partial pressure increased significantly in two groups (P < 0.05), but there was no significant difference between the two groups (P > 0.05). The proportion of blood transfusion in group LA was significantly lower than that in group C (χ2 = 4.902, P < 0.05). Conclusion Low central venous pressure combined with acute hypervolemic hemodilution has no significant inhibition on intrapulmonary shunt and arterial oxygen partial pressure during one lung ventilation during thoracic surgery, and can significantly reduce the proportion of intraoperative blood transfusion.

Construction and practice of education system for information management and information system major based on advantages and characteristics of Chinese medicine subject
MI Li1 WANG Zhenguo2 HE Yong1 LI Xuebo1 WANG Ruixiang1
At present, 42 medical colleges and universities in China (9 Chinese medicine colleges) have set up the information management and information system major. In the Chinese medicine colleges, unclear students training target causes the mismatching between students′ training and the development of traditional Chinese medicine (TCM). Because of the multidimensional main courses (economics, management, computer science and background courses of TCM), the curriculum system is disintegrated. At the same time, the theory of information management and information system technology has become an important support and protection in the field of TCM that calls for students with the ability of practice, application and innovation. Since the establishment of information management and information system specialty in 2006, the education idea of highlighting characteristic and innovation were put forward, and the college carried out some research and innovation in the training project, curriculum system, teaching mode, theory, practice and support platform, having improved the “three stage penetration” courses teaching mode of knowledge learning. Course practice and job practice, formed the “multi-leading” practice teaching mode such as Chinese literature, Chinese medicine information, information technology, information management. The information management and information system specialty has constructed abundent ways to strengthen the platform for innovation, forming the multi-level student training system.

Relevant theories discussion on the incidence of depression and yang qi in the five internal organs
WANG Liyuan1 ZHU Weining2 SHI Ling3
Yang qi is the foundation of human spirit life activity, which plays a crucial role in our life. The adequate yang qi ensures the orderly life metabolism. On the contrary, insufficient yang qi will weaken or decrease the function of viscera, and make the ascending and descending functional activities of qi abnormal. It will thus affect the five minds and cause a series of complex emotional diseases. As a common thymopathy, depressive disorder is classified as the category of depression in traditional Chinese medicine. It is believed that the pathogenesis of depression is closely related to yang qi, and is the product of insufficiency of yang qi and low dynamic state. Meanwhile, this article discusses that the insufficiency of the five internal organs is closely related to the occurrence of depression, and analyzes theoretically in detail. Combining with the etiology and pathogenesis, this article puts forward a basic depression treatment principle based on warming yang and regulating qi and related viscera function. Common symptoms are summarized in clinical diagnosis and treatment, so as to provide an effective basis for the clinical prevention and treatment of the disease.

Exploration and research on curriculum construction of Chinese medicine culture
FU Wendi
Traditional Chinese medicine discipline is the comprehensive summary of traditional Chinese medicine culture, which is systematic and theoretical. In current situation, traditional Chinese medicine is highly valued by our country, which speeds up the college education development of Chinese medicine that regards Chinese medicine culture as the breakthrough and brings about great opportunities for Chinese medicine culture. Although Chinese medicine culture discipline develops in a fast way, it is in the spontaneous and free construction stage, which has no consensus about some basic problems in Chinese medicine culture. The construction of Chinese medicine culture discipline should be considered from the aspects of theory construction, practice exploration and future development. Its basic theory, course exploitation, technology integration and method creation should be paid more attention. Guiding ideology, basic content, teaching method and education for teachers should be the breakthrough, which makes the course construction of Chinese medicine culture be the pioneer of Chinese medicine education reform. It contributes to the cultural heritage of Chinese medicine.

Objective To analysis the trend and characteristics of hospitalization expenses structure change in a grade Ⅲ level A general hospital, and provide evidence for the unreasonable increase of hospitalization expenses. Methors The hospitalization expenses of patients in a grade Ⅲ level A general hospital from 2014 to 2016 were collected by analyzing the structure changes value, degree of structure variation and structure contribution rate. Results The total hospitalization expenses of the top three hospitals increased year by year from 2014 to 2016, but the growth rate decreased. The proportion of medical expenses and medical materials expenses accounted for the total hospitalization expenses of top 2. The value of negative changes in structure of 2014-2016 medicine fees, examination fees, examination fees, bed charges, treatment fees and nursing; value positive changes in the structure of health laboratory fees, material fee, operation fee and other expenses. The cost of medicine and medical materials contribute greatly to the change of hospitalization expenses. The cumulative contribution rate of these 2 items was 47.06%. Conclusion Expenses control policies for medicine has achieved initial success, but the proportion of medicine expenses is still high which still need to strengthen the medicine expenses control. The structure of hospitalization expenses is unreasonable. The proportion of medical materials and laboratory fees are high. However, the cost of operation and nursing care which reflects the technical labor value of medical staff is relatively low. Expenses structure needs to be adjusted and optimized.
