LIU Deliang1 LI Huilin1 QU Xin1 ZHAO Hengxia1 CHU Shufang1 LIU Xuemei1 ZENG Lin2 ZHANG Xuewen2
Objective To investigate the preventive effect of Huoxue Jiangtang Decoction on pancreatic β cell apoptosis and its influence on scrib protein expression in type 2 diabetes mellitus rats. Methods The type 2 diabetes mellitus model was induced by high fat and high calorie diet and intravenous injection of low dose of Streptozotocin. By SPSS software, all model rats were divided into model group, herbal group, Metformin group, besides, normal group was added, 10 rats in each group. After treatment, the blood glucose, insulin and lipids were tested; the pancreatic β cell apoptosis was examined by terminal-deoxynucleoitidyl transferase mediated nick end labeling method; and scrib protein expression was detected by Western blot in extracted and purified pancreatic islet cells. Results Compared with model group, the levels of fasting blood-glucose, 2 h postprandial blood glucose, fasting insulin, 2 h postprandial insulin, insulin resistance index and total cholesterol, triacylglycerol, low density lipoprotein cholesterin in herbal group and Metformin group were all decreased, the differences were statistically significant (P < 0.05 or P < 0.01), while high density lipoprotein cholesterol had no obvious changes, the difference was not statistically significant (P > 0.05). Compared with model group, the β cell apoptosis indices in herbal group and Metformin group were also decreased (P < 0.01), meanwhile, the scrib protein expression in islet cells was increased significantly (P < 0.01). Conclusion Huoxue Jiangtang Decoction treatment has the effects of regulating blood lipid, decreasing blood glucose, improving insulin resistance, which can effectively prevent β cell apoptosis. The mechanism may be related to the upregulation of scrib protein expression in islet cells.

FENG Yue1,2 ZHANG Zongpeng1,2 SHEN Xiuping1,2 HU Jinfang1,2
Objective To study the analgesic effect of Yuanhu Zhitong Dropping Pills on dysmenorrhea model rats and to discuss the possible mechanism. Methods Sixty female SPF grade Wistar rats were divided into control group, model group, small-dose (0.2 g crude drug/kg), low-dose (0.4 g crude drug/kg), medium-dose(0.8 g crude drug/kg), high-dose(1.6 g crude drug/kg) of Yuanhu Zhitong Dropping Pills group according to weight random block method, with 10 rats in each group. The model of dysmenorrhea was induced by intraperitoneal injection of oxytocin, and Yuanhu Zhitong Dropping Pills was administered by intragastric administration for 3 days after sensitized by intraperitoneal injection of estradiol benzoate. The effect of Yuanhu Zhitong Dropping Pills was observed by detecting latency stage and times of writhing response induced by dysmenorrhea, and the level of factors related to pain such as prostaglandin F2α and prostaglandin E2 in plasma were also detected. Results Compared with model group, the times of writhing response in low-dose , medium-dose, high-dose of Yuanhu Zhitong Dropping Pills group significantly decreased (P < 0.05 or P < 0.01), and the preclinical period of writhing response in low-dose, medium-dose, high-dose of Yuanhu Zhitong Dropping Pills group showed an extension trend, while the standard deviation was large due to individual differences, the differences were not statistically significant (P > 0.05). Compared with model group, the level of β-endorphin and prostaglandin E2 in medium-dose and high-dose of Yuanhu Zhitong Dropping Pills group increased significantly (P < 0.05 or P < 0.01), and the level of noradrenaline, 5-hydroxytryptamine, the ratio of prostaglandin F2α to prosta-glandin E2 decreased significantly (P < 0.05 or P < 0.01), the level of prostaglandin F2α in high-dose of Yuanhu Zhitong Dropping Pills group decreased significantly (P < 0.05). There was no statistically significant difference on the level of thromboxane B2 and 6-Keto-prostaglandin F1α between medium-dose, high-dose of Yuanhu Zhitong Dropping Pills group and model group (P > 0.05). Conclusion Yuanhu Zhitong Dropping Pills has significant analgesic effect on dysmenorrhea model rats with the dose of 0.8-1.6 g crude drug/kg. The multi-way analgesic effect is probably related to improved levels of pain-associated prostaglandins, monoamine transmitters, and β-endorphin in peripheral blood.

WANG Guanghuan1 CHEN Kaihong1 ZHONG Jun1 WANG Fusheng1 XU Chengbin1 JIANG Xuewu2
Objective To explore the effect of diethylstilbestrol (DES) on Notch signaling pathway of chorda gubernaculum in offspring rats. Methods Forty-five healthy female SD rats aged 8-10 weeks were divided into the control group, experimental group 1 and 2 according to the random number table, with 15 rats in each group. The experimental group 1 and 2 were given DES 0.1 μg/(kg·d) and DES 25 μg/(kg·d) respectively. According to the conventional ratio of male to female 2∶1 cage. The day of offspring birth of each group was recorded as day 0 after birth (PD0). The chorda gubernaculum was taken at PD1 and PD7 respectively. The expressions of Notch1, Hes1, EGFR and TGF-α were analyzed by RT-PCR and Western blot imprinting. Results At PD1 and PD7, the mRNA expression of Notch1, Hes1, EGFR and TGF-α in the experimental group 1 were higher than those in the control group and the experimental group 2 (P < 0.05), while only TGF-α mRNA in the experimental group 2 was higher than that in the control group (P < 0.05). At PD1, the levels of Notch1, Hes1 and TGF-α protein in the experimental group 1 were higher than those in the control group, and the levels of Hes1 and EGFR at PD7 were increased (P < 0.05). Compared with control group, only the levels of Notch1, EGFR and Hes1 protein were in the experimental group 2 increased at PD1, and the levels of Notch1, Hes1 and EGFR protein were increased at PD7 (P < 0.05). Compared with the experimental group 1, only EGFR protein increased in the experimental group 2 at PD1, while Notch1, Hes1 and TGF-α protein increased at PD7 (P < 0.05). Conclusion Environmental estrogen can affect the expression of Notch signaling in gubernaculum of offspring rats, which may be one of the mechanisms of exogenous estrogen affecting the development and proliferation of gubernaculum.

21 Progress in the new approaches to regulation of muscle synthesis and metabolism
REN Guangxu1 YUAN Chao2
Skeletal muscle plays an important role in human health and disease. In many tumors, chronic heart failure and nutritional deficiency, the symptoms of muscular atrophy are characterized by loss of muscle mass and strength. Muscle atrophy seriously affects the quality of life, and even threatens the lives of patients. Although researchers have found that inflammation and nutritional health state can lead to muscle atrophy, the specific regulatory network is still not clear. Recent studies have revealed that gut microbes in the human body may also participate in the regulation of muscle synthesis. Therefore, we will review the recent progress in the study of the "gut muscle" axis and the molecular mechanisms involved.

25 General experiment study about regulating effect on OPG/RANK/RANKL system of ovariectomized osteoporosis rats by traditional Chinese medicine and acupuncture
ZHOU Jingying1 ZHOU Ning2 ZHANG Yiweng3 YANG Hongguang4 ZHAO Yingxia3
The pathogenesis of postmenopausal osteoporosis is closely related to the dysfunction of osteoprotegerin (OPG)/ receptor activator of nuclear factor κB (RANK)/ receptor activator of nuclear factor κB ligand (RANKL) system. Immature osteoblast secretes RANKL, which is combined with RANK, then it will stimulate the differentiation of osteoclast and inhibit its apoptosis, promoting bone absorption. Mature osteoblast secretes OPG, which will competitively combine RANKL with RANK, thus inhibit osteoclast differentiation and induce its apoptosis, reducing bone resorption. OPG/RANK/RANKL system can be regulated by traditional Chinese medicine and acupuncture, especially the traditional Chinese medicine and acupoints of tonifying kidney and spleen. This may be one of the mechanisms of traditional Chinese medicine and acupoints to prevent and treat postmenopausal osteoporosis.

36 The research progress on the pain treatment with Vitamin C
LIU Yao1 XIONG Miaomiao1 CHEN Yajie1 JIN Jun1 SHEN Wen2▲
Vitamin C (VitC), also known as ascorbic acid, is a highly effective antioxidant in the human body and plays an important role in many biosynthetic processes. VitC deficiency is highly prevalent in a variety of patients, such as patients with trauma, infection, and cancer. Many recent clinical studies have shown that intravenous high-dose VitC can prevent the occurrence of complex regional pain syndrome after fracture. It can also relieve the herpes zoster pain and reduce the incidence of postherpetic neuralgia. In addition, the high-dose VitC can reduce pain in patients with advanced cancer and extend their survival time. Many experts have proposed several mechanisms for relieving pain in VitC, but they are still controversial. All in all, VitC has shown a safe and effective effect in relieving acute and chronic pain.

SU Yang ZHANG Qian CUI Ronggang LI Juan TIAN Fengsheng
Objective To study the risk and influencing factors of thrombosis in patients with early diabetic nephropathy. Methods One hundred and twenty patients with type 2 diabetes that admitted to Endocrinology Department of Cangzhou Hospital of Integrated Chinese and Western Medicine("our hospital" for short) from May 2017 to May 2018 were selected, with 60 cases ACR<2.5 (male)/3.5 (female) mg/mmol as type 2 diabetes group (T2DM group), 60 cases ACR from 2.5 (male)/3.5 (female) mg/mmol to 30.0 mg/mmol as early diabetic nephropathy group (EDN group). And 30 cases of health check-up in our hospital were taken as the control group(NC group). The metabolism indexes of each group were detected as fasting blood-glucose (FPG), postprandial 2 h plasma glucose (2hPBG), glycosylated hemoglobin (HbA1c), total cholestero (TC), low-density lipoprotein cholesterol (LDL-C), triacylglycerol (TG), and thrombelastogram (TEG) indexes as R value, K value, CI value, MA value, Angle angle value to assess the risk of thrombosis. Results Compared with the NC group, the levels of FPG, 2hPBG, HbA1c of T2DM group and EDN group increased (P < 0.05); compared with the T2DM group, the levels of FPG, 2hPBG, HbA1c of EDN group increased (P < 0.05). Compared with the NC group, the levels of TG, TC, LDL-C of T2DM group and EDN group increased (P < 0.05), while there was no significant difference in the levels of TG, TC, LDL-C between T2DM group and EDN group (P > 0.05). Compared with the NC group, the R value and K value of T2DM group and EDN group decreased, while the Angle value, MA value and CI value increased, and the differences were statistically significant (P < 0.05); compared with the T2DM group, the R value and K value of the EDN group decreased, while the Angle value, MA value and CI value increased, the differences were statistically significant (P < 0.05). Linear correlation analysis showed that ACR was negatively correlated with R value and K value (r = -0.423, -0.517, all P < 0.01), ACR was positively correlated with Angle value, MA value and CI value (r = 0.683, 0.742, 0.669, all P < 0.01). Logistic regression analysis showed that the course of diabetes, dyslipidemia, smoking history, and ACR increase were independent risk factors for thrombosis in patients with early diabetic nephropathy (OR = 2.423, 1.986, 2.103, 4.358, all P < 0.05). Conclusion Diabetics have a higher risk of blood clots than healthy adults before proteinuria. Patients with early diabetic nephropathy have a higher risk of thrombosis than those with simple diabetes. 98 Experience of treating chronic kidney diseases from the Shanghai style famous doctor TONG Shaobo
WANG Jie CHEN Wenhao SUN Beibei JI Jing LI Yuting YU Hongji HE Liqun
TONG Shaobo is a famous doctor of Chinese medicine and the successor of Ding′s internal medicine of Shanghai. In the long term clinical work, TONG Shaobo proposes to grasp the following key points in treating chronic kidney disease: Firstly, it is important to adjust the Yin and Yang, and give consideration of the lung, spleen and kidney. Secondly, the outside and the inside of body should be paid attention to, and adjust the tri-jiao. Thirdly, it is essential to strengthen the body resistance to eliminate pathogenic factors. TONG Shaobo believes that chronic kidney disease is the evidence of the deficiency, most of which are integrated false and solid, especially the deficiency of the spleen and kidney is the basis, leading to the damp wet, phlegm, blood stasis and other solid pathogenic factors. These factors will in turn lead to the dysfunction of the body, and which is difficult to be cured. Therefore, TONG Shaobo formulates a series of rules of treatment, taking into account the three organs-lung, spleen and kidney, paying attention to the harmonization of Yin and Yang. His treating rules not only helps the positive Qi, but also takes into account the existence of the pathogenic factors. He formula a series of therapies and rules in treatment, and uses multiple methods according to different syndrome differentiation.
