Study of anti-inflammatory effect and molecular mechanism of Yinyanjing Lotion |

Abstract Objective To investigate anti-inflammatory effect and molecular mechanism of Yinyanjing Lotion on allergic contact dermatitis (ACD) rats. Methods Fifty half male and half female SPF grade SD rats were randomly divided into 5 groups according to random number table method, including blank control group (purified water 50 μL), ACD model group (purified water 50 μL), positive control group (Dexamethasone Sodium Phosphate Injection 50 μL), low dose treatment group (Yinyanjing Lotion 50 μL) and high dose treatment group (2 times concentrated Yinyanjing Lotion 50 μL), with 10 rats in each group. The right ears were smeared with drugs twice a day for 10 days. The allergic reaction and swelling degree of the skin were observed, and the expression levels of phosphorylated extracellular regulated protein kinases (pERK1/2) and nuclear factor κB (NF-κB) were detected by immunohistochemistry and Western blot. Results Compared with the blank control group, the ear swelling degree of rats in the ACD model group was significantly increased (P < 0.01). Compared with the ACD model group, the ear swelling degree of rats in the high dose treatment group and low dose treatment group was significantly decreased (P < 0.01). Compared with the blank control group, the protein expression of pERK1/2 and NF-κB in the ACD model group was significantly up-regulated (P < 0.01). Compared with the ACD model group, the protein expression of pERK1/2 and NF-κB in the high dose treatment group and low dose treatment group was significantly down-regulated (P < 0.05 or P < 0.01). Conclusion Yinyanjing Lotion has an anti-inflammatory effect on ACD rats, and the mechanism may be achieved by inhibition of pERK1/2/NF-κB signaling pathway.
