Abstract  Objective Qualitative and quantitative analysis of the chemical constituents of flavonoids in the leaves of Tetrastigma hemsleyanum. Methods The chemical constituents in leaves of Tetrastigma hemsleyanum were identified by ultra performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS). The column was C18 Luna Omega (2.1 mm × 100 mm, 1.6 μm); the mobile phase was acetonitrile-water (50∶50, V/V); the flow rate was 0.4 mL/min; the column temperature was 35°C; the injection volume was 5 μL; full scan mode. The contents of five carbon glycosides in the leaves of Tetrastigma hemsleyanum were determined at the same time by high performance liquid chromatography (HPLC). The column was Ultimate XB C18 (250 mm×4.6 mm, 5 μm); the mobile phase was acetonitrile - 0.2% H3PO4 (10∶90, V/V); the flow rate was 1 mL/min; the detection wavelength was 350 nm; room temperature; injection volume was 20 μL. Results A total of 6 chemical components were identified in the leaves, and the retention time, spectrum and mass spectrometry information of the 5 flavonoids were consistent with the control, which were isohumullin, valerin, vitexin and rhamnoside, vitexin, isovite. The contents of the above five components in the samples from 8 different origin sources were determined by HPLC. The linear relationship of each component was good (r > 0.9997), and the recovery rate was between 95.0% and 105.0%. Conclusion The leaves of Tetrastigma hemsleyanum are mainly composed of two kinds of carbon flavonoids, valerin and vitexin. The method established in this study can qualitatively and quantitatively analyze the main chemical components in the leaves of Tetrastigma hemsleyanum and control the quality of the medicinal materials.
