Study on the anesthetic effect of new type topical anesthetic on oral mucosa surface |

Abstract Objective To investigate the anesthetic effect of new type topical anesthetic which is Lidocaine hydrochloride lysine-linoleic acid modified dextran polymeric lipid vesicles on oral mucosa surface. Methods Eighteen New Zealand rabbits (male or female, body weight [2.0±0.2] kg) aged eight months were divided into negative control group (0.5 ml of Lidocaine hydrochloride injection was applied on the labial gingival of the lower anterior teeth),positive control group (0.5 ml of Lidocaine hydrochloride conventional liposomes was applied on the labial gingival of the lower anterior teeth) and experimental group (0.5 ml of Lidocaine hydrochloride lysine-linoleic acid modified dextran polymeric lipid vesicles was applied on the labial gingival of the lower anterior teeth) by stratified random grouping according to body weight, with six rats in each group. Acupuncture experiment and mental nerve action potential experiment were performed in the three groups, and the experimental results were compared. Results The results of acupuncture experiment showed that the onset time of anesthesia in the experimental group was shorter than that in the positive control group, and the difference was statistically significant (all P < 0.05). No significant anesthetic effect was found in negative control group. Overall analysis of the inhibition rate of mental nerve action potential amplitude (APA) among the three groups showed that there were statistically significant differences among groups, time point comparison and interaction (P < 0.05). Comparison within groups: the inhibition rate of APA at 50 min in negative control group and positive control group was higher than that at other time points, with statistical significance (all P < 0.05); the inhibition rate of APA in experimental group at 40 min was higher than that at other time points, with statistical significance (all P < 0.05). Comparison between groups: the APA inhibition rate of the experimental group was higher than that of the positive control group at each time point, and the APA inhibition rate of the positive control group was higher than that of the negative control group within 50 min, with statistical significance (all P < 0.05). Conclusion Lidocaine hydrochloride lysine-linoleic acid modified dextran polymeric lipid vesicles has a quick effect and good anesthetic effect, which has a good clinical application prospect.
