Determination of the content of five components in crossbow liquid by HPLC |

Abstract Objective To establish a high performance liquid chromatography (HPLC) for the determination of five components of anabasine, catechin, chlorogenic acid, mesaconitine and hypoaconitine in crossbow liquid. Methods Chromatographic column Agilent 5 TC-C18(2) (250.0 mm×4.6 mm, 5 μm) was used, with acetonitrile-0.2% phosphoric acid aqueous solution as the mobile phase, gradient elution, flow rate was 1.0 mL/min, column temperature was 25℃ and 30℃, the detection wavelengths were 258 nm and 235 nm, and the injection volume was 10 μL. Results The anabasine, catechin, chlorogenic acid, mesaconitine and hypoaconitine showed good linear relationship in the range of 2.797-14.161, 25.816-130.694, 7.956-40.275, 2.675-13.540, 1.285-6.504 μg/mL respectively(r ≥ 0.9996); the average recovery rate was between 99.374% and 101.559%, and the RSD was between 1.144% and 2.844%. Conclusion The method is fast, efficient, sensitive and has a wide range of applications. It can provide a theoretical basis for the clinical efficacy of crossbow liquid solution.
