Abstract Objective To investigate the expression and correlation of human β-defensin-3 (hBD-3) and nuclear factor kappaB (NF-κB) p65 in normal oral mucosa, oral potentially malignant disorder (OPMD), and oral squamous cell carcinoma (OSCC). Methods Clinical data and histological specimen from the Department of Pathology, Haikou Affiliated Hospital of Center South University Xiangya School of Medicine from January 2015 to December 2018, 60 patients with OPMD, 40 patients with OSCC and 15 cases of normal oral mucosa were collected. Immunohistochemical staining was applied to investigate the expression of hBD-3 and NF-κB p65 in paraffin sections of normal oral mucosa, OPMD, and OSCC tissues. The expression in different types of tissue specimens and their correlation were analyzed. Results The expression intensity of hBD-3 and NF-κB p65 in OSCC was significantly higher than those in normal group and OPMD group (P < 0.05), and OPMD group was significantly higher than that in normal group (P < 0.05). The expression of hBD-3 was positively correlated with NF-κB p65 (r = 0.355,P < 0.05). HbD-3 and NF-κB p65 were associated with OPMD epithelial dysplasia (P < 0.05). Conclusion The expression of hBD-3 and NF-κB p65 protein is increased in the early carcinogenesis of oral mucosa and has obvious synergistic effect, suggesting that they may be involved in the occurrence and development of oral squamous cell carcinoma.
