Abstract Objective To investigate the expression, clinical significance and correlation of chemokine receptor 2 (CXCR2) and growth regulatory gene α (GROα) in esophageal squamous cell carcinoma. Methods Seventy cases of esophageal squamous cell carcinoma and paracancerous normal tissues were selected from the thoracic surgery department of the First Affiliated Hospital of Hebei North University from January 2017 to June 2018. The expression of CXCR2 mRNA and GROα mRNA in the two groups were detected by reverse transcription-polymerase chain reaction (qRT-PCR). Immunohistochemistry (IHC) was used to detect the expression of CXCR2 and GROα protein in the two groups. At the same time, the clinical data of the patients were collected to analyze the relationship between the expression of the two and the clinicopathological characteristics, Spearman test was used to analyze the correlation between the two proteins. Results The qRT-PCR results showed that the expression level of CXCR2 mRNA in cancer tissue was significantly higher than that in normal tissue, and the difference was highly statistically significant (P < 0.01). The expression level of GROα mRNA in cancer tissue was significantly higher than that in normal tissue, and the difference was highly statistically significant (P < 0.01). The results of IHC method showed that the positive expression rates of CXCR2 and GROα in the cancer tissue were significantly higher than those in the normal group, and the difference was highly statistically significant (P < 0.01). The expression levels of CXCR2 and GROα in the cancer tissue were correlated with the degree of differentiation, TNM stage, lymph node metastasis and liver metastasis (all P < 0.05). And not related to tumor size (P > 0.05), Spearmen correlation analysis showed that CXCR2 was positively correlated with GROα in cancer tissues (r = 0.476, P < 0.001). Conclusion Abnormal expression of CXCR2 and GROα plays an important role in the occurrence and development of esophageal squamous cell carcinoma. Combined detection of the expression levels of CXCR2 and GROα in esophageal squamous cell carcinoma can be used as a potential molecular biological indicator for early diagnosis and treatment monitoring of esophageal squamous cell carcinoma.
