Study on the blood flow effect of Invigorating Spleen, Benefiting Qi, Controlling Blood Formula and its decomposed recipes on Simvastatin-induced zebrafish cardiac hemorrhage model |

Abstract Objective To observe the effect of Invigorating Spleen, Benefiting Qi, Controlling Blood Formula on the blood flow of Simvastatin-induced zebrafish cardiac hemorrhage model and to explore the relationship among the hemodynamic changes of zebrafish by different treatment. Methods The fertilized egg of 660 72-hour Albino zebrafish in melanin allele mutation was randomly divided into the normal control group, the model control group and group A (Invigorating Spleen Granule Formulations), B (Invigorating Spleen, Benefiting Qi Granule Formulations), C (Invigorating Spleen, Benefiting Qi, Replenishing Blood Granule Formulations), D (Invigorating Spleen, Benefiting Qi, Replenishing Blood, Controlling Blood Granule Formulations) with different concentrations (100, 250, 500, 750, 1000 μg/mL) of experimental drugs. Each group of 30. First, the zebrafish of group A, B, C, D were treated with different concentrations of drugs for 4 hours, and then with 50 μmol/L Simvastatin for 30 minutes. Next, through collecting blood flow imaging of the 20 zebrafish in each group by heartbeat blood flow analysis system, the blood flow was analyzed and the improvement rate was calculated. Results ①Compared with the model control group, different concentrations of drug groups in group A, B had no effects on the improvement of blood flow in Simvastatin-induced zebrafish hemorrhage model; however, 500, 750,1000 μg/mL of drugs in group C, D could significantly increase the improvement rate of blood flow in zebrafish hemorrhage model (P < 0.05). ②In ameliorating the model's velocity of blood flow and its improvement rate, 1000 μg/mL of Invigorating Spleen, Benefiting Qi, Replenishing Blood, Controlling Blood Formula was superior to that of Invigorating Spleen, Benefiting Qi, Replenishing Blood Formula. Conclusion The comprehensive application of invigorating spleen, benefiting qi, replenishing blood, controlling blood can ameliorate the blood flow of Simvastatin-induced zebrafish cardiac hemorrhage model, which reflects the theory of “qi is the commander of blood and blood is the mother of qi”.
