Screening of active parts of Simiao Yong′an Decoction based on PPAR-γ agonist and analysis its chemical composition |

Abstract Objective To screen the active parts of Simiao Yong′an Decoction with peroxisome proliferators-activated receptor γ (PPAR-γ) activation and analyze its chemical components, and to explore the active components and anti-inflammatory effects of the extract. Methods Eight alcohol extracts of Simiao Yong′an Decoction were obtained by alcohol extraction and resin separation. The PPAR-γ was used as the target to screen the active parts of the extract of Simiao Yong′an Decoction at the cell level. A dose-dependent curve was drawn for the extracts with active tendency. EC50 value and relative activity value were calculated, and the chemical composition of active parts was analyzed and identified by liquid chromatograph-tandem mass spectrometer technology. Results In the PPAR-γ activation model, the extracts CS04 and CS05 have strong PPAR-γ activation. The chemical components of the active parts CS04 and CS05 with strong PPAR-γ agonistic effects were identified. A total of 31 components were identified. Conclusion CS04 and CS05 of Simiao Yong′an Decoction have strong PPAR-γ activation, which may be related to the inhibition of atherosclerosis inflammation.
