Simultaneous determination of Prim-O-Glucnylcimifugin and 4′-O-β-glucosyl-5-O-Methylvismmiside in Compound Jingfang Granules by SPE-UPLC |

Abstract Objective To establish a solid phase extraction and ultra performance liquid chromatography(SPE-UPLC) method for the simultaneous determination of Prim-O-Glucnylcimifugin and 4′-O-β-glucosyl-5-O-Methylvismmiside in Compound Jingfang Granules. Methods Samples were processed by the solid phase extraction system, and the separation was performed on ACQUITY UPLC HSS T3 (2.1 mm×100 mm,1.8 μm) with acetonitrile-water mobile phase for the gradient elution. The flow rate was 0.4 mL/min, the detection wavelength was 254nm, and the column temperature was 35℃. Results The calibration curve for Prim-O-Glucnylcimifugin and 4′-O-β-glucosyl-5-O-Methylvismmiside in the range of 5-80 mg/L showed good liner. The linear regression equation for Prim-O-Glucnylcimifugin were A = 40 195C-248.67 (r =1.0000) and the linear regression equation for 4′-O-β-glucosyl-5-O-Methylvismmiside were A = 42 577C+12 782(r =1.0000). The average recovery of Prim-O-Glucnylcimifugin and 4′-O-β-glucosyl-5-O-Methylvismmiside were 98.11% and 99.28%, and RSD were 1.92% and 1.90%(n = 6), respectively. Conclusion SPE-UPLC method is simple, accurate and reproducible and effectivly control the quality of Compound Jingfang Granules.
