Simultaneous determination of two constituents in Chuangui Bushen Tongluo Capsules by high performance liquid chromatography |

Abstract Objective To establish a high performance liquid chromatography (HPLC) method for simultaneous determination of salvianolic acid B and tanshinone ⅡA in Chuangui Bushen Tongluo Capsules. Methods The content of salvianolic acid B and tanshinone ⅡA was determined by HPLC. Agilent ZORBAX SB-C18 (5 μm, 4.6 mm×250 mm) was used as the column. Acetonitrile(A)-0.1% aqueous phosphoric acid (B) was used as the mobile phase with gradient elution. With the detection wavelength of 270 nm tanshinone ⅡA and 286 nm vianolic acid B. The flow rate was 1.0 mL/min and column temperature was 20℃. Results The linear ranges of salvianolic acid B and tanshinone ⅡA were 44.83-448.3 μg/mL, 0.96-9.60 μg/mL, respectively. The regression equation were Y = 12 650X-1973.3(r = 0.9996),Y = 54 274X+22 206 (r = 0.997), respectively. The average recoveries were 99.7% (RSD = 1.9%), 97.4% (RSD = 1.7%), respectively. Conclusion The established method has the advantages of simple operation, strong specificity and good reproducibility, which can be used to control the quality of Chuangui Bushen Tongluo Capsules.
