Expression and significance of IL-17 and NF-κB in the rats with acute gout arthritis |

Abstract Objective To investigate the expression and significance of interleukin-17 (IL-17) and nuclear factor kappa-B (NF-κB) in the acute gouty arthritis (AGA) rat model. Methods A total of 45 SD rats were divided into control group (9 rats) and model group (including four subgroups: 6, 12, 24 h and 72 h, 9 rats in each group) by random number table. A rat model of AGA was induced with the injection of monosodium urate (MSU) into the right posterior ankle joint cavity. The rats of the control group were injected with an equal dose of saline. The model group was sacrificed at 6, 12, 24 h and 72 h, respectively. The expression of interleukin-1β (IL-1β), tumor necrosis factor-α (TNF-α) and interleukin-17 (IL-17) in serum and synovium was detected by the enzyme-linked immunosorbent assay. The expression of NF-κB in synovial membrane was detected by Western blot. Results Compared with the control group, the expression of IL-1β, TNF-α and IL-17 in the model group was significantly increased at 6 h after AGA (P < 0.01), peaked at 12 h, and began to decrease at 24 h. The relative expression level of p-NF-κB p65 in the synovial membrane of the model group was significantly higher than that of the control group, and gradually decreased with time. Conclusion After AGA, the expression levels of IL-17 and p-NF-κB p65 are significantly increased, and the inflammatory response is aggravated. The differential expression of IL-17 and NF-κB is closely related to the occurrence and development of AGA.
