Differential expression of microRNA in atypical endometrial hyperplasia based on high-throughput sequencing |

Abstract Objective To investigate the differentially expressed microRNAs (miRNAs) and significance in endometrium from atypical endometrial hyperplasia (AEH) and normal women through high-throughput sequencing. Methods From January 2016 to January 2017, three patients diagnosed with AEH (experimental group) and three normal women (control group) admitted to Beijing Obstetrics and Gynecology Hospital, Capital Medical University were collected. Second-generation high-throughput sequencing was used to screen the differentially expressed miRNAs. Databases was used to predict its target genes and biological functions, and understand the closely related miRNAs and their co-regulated target genes. Results There were 18 differentially expressed miRNAs in two groups, which 17 were up-regulated and 1 was down-regulated. Most of the target genes with differential miRNA were concentrated in the signal transduction pathways related to cancer or body metabolism. Among the differentially expressed miRNAs, miR-577, miR-182-5p and miR-4286 are closely linked. They co-regulate the expressions of CACNA1C, MECP2, FOXK1, LPP, SH3TC2, Smad and MDM4. Conclusion There are significantly differential expressions of miRNAs in endometrium from AEH and Normal endometrial tissue. These miRNAs may be involved in the occurrence and development of AEH via adjusting different target genes to regulate the signal pathway.
