Effect of naringin and naringenin on Clarithromycin metabolism in rat liver microsomes |

Abstract Objective To investigate the effect of naringin and naringenin on Clarithromycin metabolism in rat liver microsomes. Methods The content of Clarithromycin in rat liver microsomes was determined by high performance liquid chromatography. The Michaelis constant (Km) for Clarithromycin was calculated according to Lineweaver-Burk equation. The effect of naringin and naringenin at concentrations of 0.11-180.00 μmol/L on Clarithromycin metabolism were evaluated in rat liver microsome by incubation in vitro. Results The chromatograms showed that Clarithromycin and inter standard were separated well and both were not disturbed by endogenesis substances, there was no interference of other endogenous substances in the warm incubation system. Vmax and Km were 0.59 nmol/(min·mg) and 77.57 μmol/L, respectively. Compared with the blank control group, naringin and naringenin in each dose group showed no statistically significant difference in Clarithromycin metabolism in rat liver microsomes (P > 0.05). Conclusion Naringin and naringenin did not affect the metabolism of Clarithromycin by liver microsomes.
