Effect of 2-methoxyestradiol on proliferation, migration and radioresistance of nasopharyngeal carcinoma stem cells |

Abstract Objective To investigate the effect of 2- methoxyestradiol (2-ME2) on proliferation, migration and resistance to radiotherapy in nasopharyngeal carcinoma stem cells. Methods Nasopharyngeal carcinoma CNE-2 cells were selected to construct nasopharyngeal stem cell model, they were randomly divided into 0 μmol/L group, 4 μmol/L group and 8 μmol/L group by using random number table method. The proliferation of cells in each group was detected by CCK-8 cell proliferation test, Transwell cell migration test was used to detect the cell migration in each group, clonogenic assay was used to detect cell radiotherapy resistance, Western blot method was used to detect the expression of nuclear factor-k-gene-binding (NF-κB) p65 and hypoxia inducible factor-1α (HIF-1α) protein. Results OD value of cells in 4 μmol/L group and 8 μmol/L group was significantly lower than that in 0 μmol/L group (P < 0.05), and that in 8 μmol/L group was significantly lower than that in 4 μmol/L group (P < 0.05). The number of perforating cells in 4 μmol/L group and 8 μmol/L group was significantly less than that in 0 μmol/L group (P < 0.05); the number of perforating cells in 8 μmol/L group was significantly less than that in 4 μmol/L group (P < 0.05). The colony-forming ability of cells in 4 μmol/L group and 8 μmol/L group after 0 and 8 Gy irradiation was significantly lower than that in 0 μmol/L group (P < 0.05); the colony-forming ability of cells in 8 μmol/L group after 0 and 8 Gy irradiation was significantly lower than that in 4 μmol/L group (P < 0.05). The expression of NF-κB p65 and HIF-1α in 4 μmol/L group and 8 μmol/L group was significantly lower than that in 0 μmol/L group (P < 0.05); the expression of NF-κB p65 and HIF-1α in 8 μmol/L group was significantly lower than that in 4 μmol/L group (P < 0.05). Conclusion 2-ME2 can inhibit the proliferation and migration of nasopharyngeal carcinoma stem cells, and reduce the radiation resistance, the mechanism may be related to the inhibition of HIF-1/NF-κB signal transduction pathway, it is worthy of further study.
