Abstract Paraoxonase-1 (PON1) is a high density lipoprotein-related esterase. PON1 plays an important physiological role in human serum. Its expression and activity are mainly affected by PON1 gene single nucleotide polymorphism. The relationship between single nucleotide polymorphism and disease is not consistent with individuals or populations. The occurrence of disease is the result of combination of genetical, physical and chemical factors. The relationship and mechanism between PON1 and many diseases remain unclear. This article aims to explore the relationship between PON1 and coronary heart disease, diabetes mellitus, stroke and cancer, and the possible pathogenesis, and provide new ideas for exploring the treatment and prevention of related diseases.
