Effect of Dark Plum Sprays on radiation-induced submandibular gland damage of rats |

Abstract Objective To observe the influence of Dark Plum Sprays on radiation-induced submandibular gland damage of rats. Methods According to the random number table, a total of 30 healthy Wistar male rats aged 6-7 weeks were divided into normal group (NG), untreated group (UG), high-dose Dark Plum Sprays group (HG), medium-dose Dark Plum Sprays group (MG), low-dose Dark Plum Sprays group (LG), each group had 6 rats. The rats were given one-time 18 Gy irradiation locally on the submandibular glands. 14 days later, saliva was collected to calculate the salivary flow rate and detected the biochemical indexes. After that, submandibular glands were draw out. Morphology examination was assessed by HE staining. Results 14 days after administration, the average body weights in UG, HG, MG, LG groups were significantly lower than those in the NG group (P < 0.05). The salivary flow rate, Na+, K+, Na+/K+ in UG, HG, MG, LG groups were lower than those in the NG group, but there were no statistically significant differences (P > 0.05). The cells of the Wistar rats in the NG group were regularly arranged, and there were no obviously pathological changes. The cells in UG group presented inflammatory infiltration and the collagen fibrils demonstrated fibrosis with significant hyperplasia. The submandibular gland cells of UG, HG, MG, LG groups had radioactive damage after irradiation. However, the submandibular gland cells of HG group was less atrophy, inflammatory and more cytoplasm secretion than UG. The restoration of submandibular gland cells in HG group were better than UG, but failed to return to normal. Conclusion High-dose Dark Plum Sprays may repair the pathomorphism of submandibular gland cells after radioactive damage, but relevant mechanisms need to be further explored.
