Protective effect of naringenin pretreatment on isolated heart ischemia/reperfusion injury in rat |

Abstract Objective To investigate the protective effect of naringenin on myocardial ischemia/reperfusion injury in isolated rat hearts. Methods 56 SD rats were randomly divided into: normal control (CON) group, naringenin (NARI) of 1, 2.5, 5, 10, 20, 40 μmol/L groups. After balance perfusion for 30 min in each group, CON group was treated with 30 min global ischemia followed by 60 min reperfusion (or 120 min for determination of the infarct size). The NARI rats hearts were pretreated with 1, 2.5, 5, 10, 20, 40 μmol/L Nari perfusion for 10 min before ischemia, and then 30 min global ischemia and 60 min reperfusion (or 120 min for determine the infarct size). The left ventricular function was obseved; levels of myocardial lactate dehydrogenase, myocardial superoxide dismutase (SOD) activity and malondialdehyde (MDA) were measured; and the percentage of myocardial infarct size was calculated in each group. Results Compared with the CON group, the recovery of heart function was increased, the content of lactate dehydrogenase and the area of myocardial infarction were decreased after ischemia/reperfusion in 2.5 μmol/L and above NARI pretreatment group (P < 0.05 or P < 0.01). Compared with the control group, the SOD activity of myocardium significantly increased and the content of MDA significantly decreased in 2.5 μmol/L and above NARI pretreatment group(P < 0.05 or P < 0.01). Conclusion Naringenin has protective effect for isolated rats hearts with ischemia/reperfusion injury, its mechanism may be associated with enhanced myocardial antioxidant capacity.
