Effect of 1, 25 (OH)2D3 on apoptosis and VDR protein expression of human leukemia 6T-CEM cell line |

Abstract Objective To investigate the effect of 1, 25-dihydroxyvitamin D3 [1, 25 (OH)2D3] on apoptosis of 6T-CEM cell lines and expression of vitamin D receptor (VDR) protein human leukemia. Methods The 6T-CEM cell lines were cultured and randomly divided into group A, B, C, D. Group A, B, C were added with 1, 25 (OH)2D3 with the concentration of 10-6, 10-7, 10-8 mol/L respectively, and group D was added with DMSO. The absorbance A value and cell proliferation inhibition rate were detected by MTT method, the cell morphology was observed by AO-EB fluorescence staining, and the cell apoptosis was detected by flow cytometer, the expression of VDR mRNA and protein were detected by RT-PCR and Western blot method. Results The absorbance of group A was apparently lower than that of group B, C and D (P < 0.05); the inhibitory rate of cell proliferation of group A, B, C was A>B>C; the CEM cell apoptosis index of group D was apparently lower than that of group A, B and C (P < 0.05); the apoptosis index of group A was apparently higher than other three groups (P < 0.05); there was no statistically significant difference in VDR mRNA expression among the four groups (P > 0.05). The VDR protein expression of group A was apparently higher than that of other three groups (P < 0.05). Conclusion 1, 25 (OH)2D3 can effectively inhibit the proliferation of 6T-CEM cells, induce its apoptosis, up-regulate the VDR protein expression.
