Mechanism of persistent low back pain after intervertebral space infection |

Abstract Objective To Discus on the Staphylococcus aureus supernatant of intervertebral disc nucleus pulposus cells proteoglycans (ACAN), type Ⅱ collagen (COL Ⅱ). Methods Cultured SD rat intervertebral disc nucleus pulposus cells were isolated in vitro and treated with Staphylococcus aureus supernatant at different concentrations (0%, 5%, 10%, 15%, 20%, 25%, 30%, 35%, 40%) to simulate the course of intervertebral space infection. CCK-8 was used to detect the proliferation activity of the second generation nucleus pulposus (P2) cells. RT-PCR was used to detect the gene expression of the second generation nucleus pulposus cells. Results This study found that the supernatant fluid of Staphylococcus aureus that accelerate nucleus pulposus cells to divide, stimulate ACAN, COL Ⅱ expression decreased (P < 0.05), the supernatant fluid concentration > 20%, can significantly inhibit extracellular matrix synthesis of nucleus pulposus. Conclusion The results showed that Staphylococcus aureus supernatant could inhibit the growth of nucleus pulposus cells in a concentration dependent manner. Stimulate ACAN, COL Ⅱ expression decrease may be the cause of intervertebral infection after continuous low back pain.
