The effect of Genistein on the CREB gene expression in rat ovarian granulosa cells |

Abstract Objective To investigate the effects of Genistein on the gene expression of cAMP response element binding protein (CREB) in the rats ovarian granulosa cells and the relationship with the estrogen receptor (ER). Methods The ovarian granulosa cells from female rat were isolated and collected, incubated and treated with Genistein (0.1, 1, 5, 10, 100 μmol/L) and ER blocker ICI182, 780 (1 μmol/L) for 24 h. Then the CREB mRNA expression level was assessed by RT-PCR. Results Compared with the control group, the expression of CREB mRNA of 1, 5 and 10 μmol/L Genistein group was higher (P < 0.05 or P < 0.01). After the cells pretreated with ER blocker ICI182, 780 for 30 min, 1, 10 mol/L Genistein added and pretreated 24 h group, the expression of CREB mRNA had no significant difference compared with the 1, 10 μmol/L Genistein group (P > 0.05). Conclusion 1-10 μmol/L Genistein could up-regulate the expression of CREB gene in the rats ovarian granulosa cells. The molecular mechanism of Genistein effect on the CREB gene expression may not be mediated by ER.
