Effects of curcumin on the expression of PTP1B and IRS-2 mRNA in liver tissue of rats with type 2 diabetes mellitus |

Abstract Objective To observe the effects of curcumin on expression of protein tyrosine phosphatase 1B (PTP1B) and insulin receptor substrate-2 (IRS-2) mRNA in liver tissue of rats with type 2 diabetes mellitus induced by high fat diet. Methods The SD rats were fed with high fat diet, then the tail vein was injected with Streptozotocin (STZ), so as to make type 2 diabetes mellitus model. The model rats were divided into model group, crucumin group, Pioglitazone group, with 10 rats in each group. After 4 weeks and 8 weeks of administration, the levels of fasting blood-glucose (FBG), fasting insulin (FINS), serum total cholesterol (TC), triacylglycerol (TG), low density lipoprotein cholesterin (LDL-C), high density lipoprotein cholesterol (HDL-C) were detected, and the liver index (LI), insulin sensitivity index (ISI) were calculated. After 8 weeks of administration, the pathological changes were observed by HE staining of liver, the mRNA expression of PTP1B and IRS-2 in liver was detected by real-time PCR. Results Compared with model group, crucumin group could significantly reduce the weight and the serum levels of FBG, FINS, LDL-C of rats with type 2 diabetes mellitus (P < 0.05), increase the insulin sensitivity, improve the fatty degeneration and inflammatory reactions of liver tissue, inhibit the expression of PTP1B mRNA, and up-regulate the expression of IRS-2 mRNA (P < 0.05). Conclusion Curcumin can improve the insulin resistance of model rats with type 2 diabetes mellitus, the mechanism may be related to inhibition of PTP1B mRNA expression and up-regulation of IRS-2 mRNA expression.
