Abstract PTEN is an important tumor suppressor gene of lung cancer, which inhibits the activation of AKT and mTOR by inhibiting PIP3 production. On the one hand, it directly inhibits tumor cells. On the other hand, it weakens the supporting effect of tumor stroma on tumor cells, and inhibits the occurrence, growth, and metastasis of lung cancer. Decreased expression of PTEN gene leads to the development of drug resistance in lung cancer treatment and reflects the prognosis of patients. However, due to the existence of epigenetic and post-translational regulation, PTEN protein deletion is significantly higher than the incidence of PTEN gene mutation. Therefore, compared with PTEN gene targeting therapy drugs, improving the expression of PTEN protein in lung cancer may be a more promising approach to PTEN gene therapy for lung cancer and inhibiting drug resistance.
