Experimental study on seborrheic alopecia in animals of Sanshen Yangfa Sprays |

Abstract Objective To investigate the therapeutic effect and anti-inflammatory effct of Sanshen Yangfa Sprays on the seborrheic alopecia animal model. Methods Sixty SPF male Wistar rats and sixty SPF male Institute of Cancer Research (ICR) mice were injected with testosterone propionate for 60 consecutive days, resulting in experimental seborrheic alopecia model. According to the weight block, the rats and mice were randomly divided into normal control group, model control group, high dose (0.290 g crude drug/mL), medium dose (0.145 g crude drug/mL), low dose (0.073 g crude drug/mL) of Sanshen Yangfa Sprays group, and Minoxidil Tincture group (0.05 g/mL), with 10 animals in each group, the length of hair was measured, the hair follicles and sebaceous glands were examinated by pathologic histology. According to the weight block, 50 SPF male C57BL/6 mice were randomly divided into blank control group, high dose (0.290 g crude drug/mL), medium dose (0.145 g crude drug/mL), low dose (0.073 g crude drug/mL) of Sanshen Yangfa Sprays group, and Minoxidil Tincture group (0.05 g/mL), with 10 mice in each group, the time of mouse hair entering the growth period from the rest period, the time from entering the growth period to growing the skin surface, and the length of hair growth were observed. The model of capillary permeability of mice induced by acetic acid and the model of mouse ear swelling induced by croton oil were used to research the anti-inflammatory effect of Sanshen Yangfa Sprays. According to weight block, 50 SPF male ICR mice were randomly divided into model control group, high dose(0.290 g crude drug/mL), medium dose (0.145 g crude drug/mL), low dose (0.073 g crude drug/mL) of Sanshen Yangfa Sprays group, and Yunnan Baiyao Tincture group, with 10 mice in each group, the absorbance of peritoneal fluid and value of ear swelling were measured. Results At the 15th, 30th, 45th, 60th day of drug administration, the hair length of rats in the high, medium and low dose of Sanshen Yangfa Sprays group were longer than those in the model control group (P < 0.05); the lesion degree of dermal tissue cells, hair follicles and sebaceous glands in rats of the high and medium dose of Sanshen Yangfa Sprays group were lower than those of the model control group (P < 0.05); the number of damaged hair follicles in the rats of the high and medium dose of Sanshen Yangfa Sprays group was lower than those of the model control group (P < 0.05). At the 15th, 30th and 45th day of administration, the hair length of the mice of the high and medium dose of Sanshen Yangfa Sprays group were longer than those of the model control group (P < 0.05); the number of damaged hair follicles in the mice of the high and medium dose of Sanshen Yangfa Sprays group were significantly lower than those of the model control group (P < 0.05); the lesion degree of dermal tissue cells, hair follicles and sebaceous glands in mice of the high dose of Sanshen Yangfa Sprays group was lower than that of the model control group (P < 0.05). The time of hair follicles from resting period to growing period, from entering growing period to growing surface of mice in high and medium dose of Sanshen Yangfa Sprays group were significantly shorter than those of blank control group (P < 0.05); the hair length was significantly higher than that of the blank control group at the 15th, 30th, 45th day of administration (P < 0.05). The absorbance of peritoneal fluid of mice in the high dose of Sanshen Yangfa Sprays group was significantly lower than that in the model control group (P < 0.05). The value of ear swelling of mice in the high and medium dose of Sanshen Yangfa Sprays group was significantly lower than that in the model control group (P < 0.01). Conclusion Sanshen Yangfa Sprays have the effect of promoting hair growth and anti-inflammatory effect on sebrrheic alopecia.
