Effect of vaccum sealing drainage in C57BL/6J mice with burn sepsis |

Abstract Objective To investigate the effect of vaccum sealing drainage (VSD) on C57BL/6J mice with burn sepsis. Methods A total of 300 male C57BL/6J mice were used to establish the model of burn sepsis. Eighty burned sepsis mice successfully were divided into experimental group and control group according to random number table, 40 mice in each group. In the experimental group, the wound was disposed and treated with VSD, the negative pressure apparatus was connected, and the negative pressure was adjusted to -120 mmHg (1 mmHg=0.133 kPa). In the control group, only wound treatment was performed, and dressing was replaced every 24 h. The fluorescence intensity and blood flow of wound skin, the levels of inflammatory cytokines such as IL-1β, IL-6, IL-8, IL-10, TNF-α and so on were statistically calculated in the two groups, and the cumulative survival rate of 7 d was observed. Results Before treatment, the fluorescence intensity of wound was similar between the two groups, and the difference was not statistically significant (P > 0.05). Compared with before treatment, the fluorescence intensity of the control group increased significantly after 24 h of treatment (P < 0.05), while the fluorescence intensity of the experimental group decreased compared with before treatment (P < 0.05). The difference between the two groups was statistically significant (P < 0.05). Compared with before treatment, the control group showed a decrease on the 1st and 2nd day after treatment, and an increase on the 3rd, 5th and 7th day after treatment, there were statistically significant differences (P < 0.05 or P < 0.01). The blood flow to the wounds on the back of the experimental group gradually increased and were significantly higher than those of the control group after treatment (P < 0.05 or P < 0.01). In the experimental group, the contents of pro-inflammatory cytokines IL-1β, IL-6 and TNF-α were significantly reduced and IL-8 was significantly increased compared with the control group (all P < 0.05), while the changes of IL-10 were not significant (all P > 0.05). From day 2, the survival rates of the experimental group were significantly higher than those of the control group, with statistically significant differences (all P < 0.05). Conclusion VSD treatment for C57BL/6J mice with burn sepsis can reduce bacteria content of wounds, improve blood supply, reduce systemic inflammatory response level of sepsis and improve survival rate.
