Causality between osteoporosis and lumbar disc herniation: a Mendelian randomized study |

Abstract Objective To study the causal relationship between osteoporosis (OP) and lumbar disc herniation (LDH) by Mendelian randomization analysis. Methods A genome-wide meta-analysis sample of calcaneal bone density was obtained through genome-wide association analysis, and a total of 17 166 350 single nucleotide polmorphism(SNPs) were collected. SNP mapping between OP and LDH outcomes was screened out, and instrumental variables SNP was obtained. The causal relationship between OP and LDH were analyzed by Mendelian randomization. Results A total of 203 SNPs, including rs10022648, rs10066763, rs10130587, as included as instrumental variables. The MR-Egger regression result P=0.442324 286, indicating no horizontal pleiotropy. IVW analysis showed that P<0.05, OR=0.893 174 092 (OR_lci95, 0.823 290 84; OR_uci95, 0.968 989 231), the risk of LDH increased by 0.89 times for every one SD decrease in BMD, and the two were negatively correlated. The result of heterogeneity test was P=0.44 with no heterogeneity. Conclusion There is a causal relationship between OP represented by bone density and LDH.
