Relationship between serum lncRNA MEG3 expression and Th17/Treg balance in children with allergic asthma

Abstract  Objective To investigate the relationship between maternal long chain non-coding ribonucleic acid maternally expressed gene 3 (lncRNA MEG3) expression and T helper cell (Th) 17/ regulatory T cell (Treg) imbalance in children with allergic asthma. Methods A total of 75 children with allergic asthma admitted to the First Affiliated Hospital of Xi’an Jiaotong University (hereinafter referred to as “our hospital”) from January 2018 to January 2022 were selected as asthma group, and 49 healthy children from the Children’s health clinic of our hospital were selected as control group. The percentage of Th17 cells, the percentage of Treg cells, Th17/Treg ratio, lncRNA MEG3 expression, interleukin (IL)-17, IL-22,IL-10,transforming growth factor-β (TGF-β) levels, and forced expiratory volume in one second (FEV1) were detected, the ratio of FEV1 to forced vital capacity (FEV1/FVC), and the percentage of FEV1 to predicted value (FEV1%pred) were calculated. The correlation between lncRNA MEG3 expression and Th17/Treg and its cytokines, and lung function were analyzed. Results The expression of lncRNA MEG3, the percentage of Treg cells in asthma group were lower than those in control group, while the percentage of Th17 cells, Th17/Treg were higher than those in control group (P<0.05). The levels of IL-17 and IL-22 in asthma group were higher than those in control group, while the levels of TGF-β and IL-10 were lower than those in control group (P<0.05). The FEV1, FEV1/FVC, and FEV1%pred in asthma group were lower than those of control group (P<0.05). The expression of lncRNA MEG3 in children with asthma was positively correlated with the percentage of Treg cells, IL-10, TGF-β, FEV1, FEV1/FVC, and FEV1%pred (r=0.502, 0.415, 0.491, 0.493, 0.428, 0.497, P<0.05), and the percentage of Th17 cells, Th17/Treg, IL-17, and IL-22 were negatively correlated (r=-0.501, -0.727, -0.631, -0.452, P<0.05). Conclusion The expression of lncRNA MEG3 in peripheral blood of patients with allergic asthma is decreased, which is related to Th17/Treg imbalance and low lung function.
