Expression of SOX4 in colorectal cancer and its effect on biological behavior |

Abstract Objective To investigate the expression of sex-determining region Y-box 4 (SOX4) in colorectal cancer tissues, and to study the effects of SOX4 on the proliferation, migration, and invasion of human colorectal cancer SW480 cells. Methods A total of 40 specimens with pathologically confirmed colorectal cancer removed during surgical treatment in the First Affiliated Hospital of Hebei North University from January 2018 to January 2019 were collected. The mRNA and protein expression levels of SOX4 in tissues were analyzed by reverse transcription quantitative polymerase chain reaction (RT-qPCR) and Western blot. Human colorectal cancer SW480 cells were cultured and transfected with negative control plasmid (siRNA-NC group) and interference plasmid (siRNA-SOX4 transfection group) using lentivirus transfection technique, and blank group was set up. Cell proliferation, migration, and invasion were detected by CCK-8 assay, cell scratch assay, and Transwell assay. Results Compared with paracancer normal tissue, the mRNA and protein expression levels of SOX4 in colorectal cancer tissue were increased. There was no significant difference in the mRNA expression level of SOX4, the protein expression level of SOX4, the proliferation ability of SW480 cells, the scratch healing rate of SW480 cells, and the invasion ability of SW480 cells between blank group and siRNA-NC group (P>0.05). Compared with siRNA-NC group, the mRNA expression level of SOX4, the protein expression level of SOX4, the proliferation ability of SW480 cells, the scratch healing rate of SW480 cells, and the invasion ability of SW480 cells were decreased in siRNA-SOX4 transfection group (P< 0.05). Conclusion The expression of SOX4 is up-regulated in colorectal cancer tissues, and the down-regulation of SOX4 expression in SW480 cells can significantly inhibit the proliferation, migration, and invasion of colorectal cancer cells.
