Analysis of effects of miR-143 regulation of TLR2 signaling pathway on ulcerative colitis based on bioinformatics |

Abstract Objective To explore the effects of miR-143 regulation of Toll-like receptor 2 (TLR2) signaling pathway on ulcerative colitis (UC) based on bioinformatics. Methods Gene data sets GSE75214, GSE53306, GSE48958 and miRNA data set GSE68306 were downloaded from GEO database for analysis. The target genes of the differential miRNAs meeting the criteria were predicted by miRDB software, and mapped with the screened differential genes, and the key miRNA in UC were selected for experiments. According to random number table method, 20 mice were divided into normal group and UC group, with 10 mice in each group. Sodium gluconosulfate was used for modeling. The pathological changes were observed by hematoxylin-eosin staining staining. The contents of tumor necrosis factor-α(TNF-α) were detected by ELISA, and the contents of TLR2 and interleukin-1β (IL-1β) in colon were detected by Western blot. The contents of miR-143, nuclear transcription factor-κB (NF-κB), and myeloid differentiation factor (MyD88) in colon tissue were determined by real-time quantitative PCR. Results There were 85 DEMs, including 34 DEMs were downregulated and 51 DEMS were up-regulated, and 19 DEMS (5 DEMS were highly expressed, and 14 DEMS were underexpressed), with miR-143 being the key miRNA. The results showed that compared with normal group, the serum TNF-α content in UC group was significantly increased, the expression of miR-143 in colon tissue was significantly decreased (P<0.05), the protein expressions of TLR2 and IL-1β were significantly increased, and the mRNA contents of NF-κB and MyD88 were significantly increased (P<0.05). Conclusion miR-143 is an important miRNA in ulcerative colitis, which may affect the development of UC through regulation of TLR2 pathway.
