Study on the apoptosis of human hepatoma cell line HepG2 induced by water extract from guiqi and tainong mango peels and its mechanism |

Abstract Objective To study the apoptotic effect and mechanism of water extract of guiqi and tainong mango peels on human hepatoma cell line HepG2. Methods Human hepatoma cell line HepG2 without being treated with water extract from mango peels was taken as control group, and human hepatoma cell line HepG2 with mango peel water extract added 4, 8, 12, 16 μg/μl, and 20 μg/μl was taken as experimental group. CCK-8 method was used to detect the inhibitory effect of water extract from guiqi and tainong mango peels on the proliferation of human hepatoma cell line HepG2, and the optimal concentration of drug action was determined; flow cytometry was used to determine the apoptosis rate of water extract from guiqi and tainong mango peels; the contents of Caspase 6 and Caspase 9 were determined by enzyme-linked immunosorbent assay. Results The optimal concentration of water extract from Guiqi mango peel on human hepatoma cell line HepG2 for 24 h was 16 μg/μl; the optimal concentration of water extract from Tainon mango peel on human hepatoma cell line HepG2 for 24 h was 8 μg/μl. The apoptosis rate of water extract from guiqi mango peel group and water extract from tainong mango peel group was higher than that of control group (P<0.05). The content of Caspase 6 of water extract from guiqi mango peel group was lower than that in control group; the contents of Caspase 9 of water extract from guiqi mango peel group and water extract from guiqi mango peel group were higher than those in control group (P<0.05). Conclusion Water extract from guiqi and tainong mango peels can promote apoptosis of human hepatoma cell line HepG2, and the mechanism may be related to the decrease of Caspase 6 and the increase of Caspase 9.
