Abstract
[Abstract] Objective To explore the effect of massage on endoplasmic reticulum stress and inflammation in skeletal muscle contusion. Methods Twenty-four 6-week-old male SPF SD rats were divided into control group, model group, massage non-acupoint group, and acupoint massage group according to the random number table method, with six rats in each group. Except the blank group, the other groups were established the models of blunt trauma of gastrocnemius. One day after modeling, massage intervention was started. The massage acupoint group was selected as yīnlíngquán, the massage non-acupoint group was selected as non-meridian and non-acupoint, and the blank group and the model group did not receive any intervention. HE staining was used to observe the morphological changes of muscle tissue. The contents of tumor necrosis factor-α (TNF-α) and Ca2+-ATPase (SERCA) were determined by enzyme-linked immunosorbent assay. The expressions of glucose-regulating protein 78 (GRP78) and calcium-reticulin (CRT) were detected by Western blot. Results Compared with blank group, the arrangement of muscle fibers in model group was more disordered, with obvious fracture, dissolution and scar tissue. Compared with blank group, the content of TNF-α in model group was increased (P<0.01). Compared with model group, TNF-α content in massage acupoint group and massage non-acupoint group was decreased (P<0.05); compared with massage non-point group, the TNF-α content of massage acupoint group was lower than that of massage non-point group (P<0.05). Compared with blank group, SERCA content in model group was decreased (P<0.05). Compared with model group, SERCA content in massage acupoint group and massage non-acupoint group was increased (P<0.05). Compared with blank group, the protein expressions of GRP78 and CRT in model group were increased (P<0.05). Compared with model group, the protein expressions of GRP78 and cRT in massage acupoint group and massage non-acupoint group were decreased (P<0.05); compared with the massage non-acupoint group, the protein expressions of GRP78 and CRT in the massage acupoint group were lower (P<0.05). Conclusion Massage can improve skeletal muscle blunt contusion and relieve endoplasmic reticulum stress. The mechanism may be related to up-regulation of SERCA and inhibition of endoplasmic reticulum stress proteins and inflammatory factors.
