Expression of lncRNAs H19 and its significance in subchondral bone of diabetic mellitus osteoarthritis |

Abstract Objective To investigate the expression of lncRNAs H19 in the subchondral bone of diabetic mellitus osteoarthritis (OA) and its significance. Methods Three patients with diabetes mellitus OA (DM-OA group), three patients with non diabetes OA (OA group) and, meanwhile, three physical examinees of the control group were selected. The expression of H19 in subchondral bone of each group was compared, H19 overexpression plasmid was constructed, and miR-141 and miR-22 mimic analogues were synthesized. The expression of β-catenin protein was detected. The differentiation function of osteoblasts was detected. Results The expression level of H19 in DM-OA group was higher than that in OA group and control group, and the expression levels of miR-141 and miR-22 in DM-OA group were lower than those in OA group and control group (P < 0.05). The expression level of H19 in OA group was higher than that in control group, and the expression levels of miR-141 and miR-22 in OA group were lower than those in control group (P < 0.05). After overexpression of H19 WT, miR-141, and miR-22 fragments, the fluorescence signal reported by luciferase decreased significantly (P < 0.01). The miR-141 mimic and miR-22 mimic analogues had no effect on the fluorescence signal of mutant plasmids, and the difference was not statistically significant (P > 0.05). The expression levels of β-catenin mRNA and Runx2 mRNA in DM-OA group were higher than those in OA group and control group (P < 0.05). The expression levels of β-catenin mRNA and Runx2 mRNA in OA group were higher than those in control group (P < 0.05). The expression levels of β-catenin protein and Runx2 protein in DM-OA group were higher than those in OA group and control group (P < 0.05). The expression levels of β-catenin protein and Runx2 protein in OA group were higher than those in control group (P < 0.05). Conclusion H19 is overexpressed in the subchondral bone of diabetes OA. H19 can activate Wnt/β-catenin signaling pathway by regulating miR-141 and miR-22 promotes subchondral bone hyperplasia and sclerosis.
