Effect of Tongqiao Yinao Prescription on learning and memory behavior in Scopolamine dementia model mice#br#

Abstract  Objective To observe the effect of Tongqiao Yinao Prescription on learning and memory behavior in Scopolamine dementia model mice. Methods A total of 50 male Kunming mice were divided into blank control group, Scopolamine model group, Donepezil group, Tongqiao Yinao Prescription 0.41 g/kg group, and Tongqiao Yinao Prescription 0.82 g/kg group by random number table method, with ten mice in each group, continuously administrate drug for 29 d. Except blank control group, other groups were intraperitoneally injected with Scopolamine Hydrobromide 2 mg/kg 30 min before behavioral experiment to establish the model, blank control group was intraperitoneally injected with normal saline. The learning and memory ability of mice was tested by platform test, the contents of acetylcholine (ACh) and acetylcholinesterase (ChAT), and the activity of acetylcholinesterase (AChE) in hippocampus tissue of each group were determined by colorimetry and enzyme-linked immunosorbent assay. Results Compared with blank control group, the incubation period of stepping down in Scopolamine model group was shortened and the number of stepping down errors was increased during the training period and test period of platform test, and the differences were statistically significant (P < 0.05); compared with Scopolamine model group, the incubation period of stepping down in Tongqiao Yinao Prescription 0.41 g/kg and Tongqiao Yinao prescription 0.82 g/kg group were prolonged and the number of stepping down errors were reduced during the training period and test period of platform test, and the differences were statistically significant (P < 0.05). Compared with blank control group, the contents of ACh and ChAT of Scopolamine model group were decreased, and the activity of AChE was increased, and the differences were statistically significant (P < 0.05); compared with Scopolamine model group, the contents of ACh and ChAT of Tongqiao Yinao Prescription 0.41 g/kg and Tongqiao Yinao prescription 0.82 g/kg group were increased, and the activity of AChE were decreased, and the differences were statistically significant (P < 0.05). There were no statistically significant differences between Tongqiao Yinao Prescription 0.41 g/kg and Tongqiao Yinao prescription 0.82 g/kg group in incubation period of stepping down, the number of stepping down errors, ACh, ChAT content, and AChE activity (P > 0.05). Conclusion Tongqiao Yinao Prescription can improve the transient acquired learning and memory impairment caused by Scopolamine to a certain extent, and its mechanism is related to regulating the cholinergic function of Scopolamine mouse model.
