Abstract Objective To investigate the relationship between human papilloma virus (HPV) infection and expressions of Toll-like receptor 4 (TLR4) and nuclear factor-kappa B (NF-κB) protein in breast cancer and its clinical significance. Methods A total of 50 cases of invasive cancer breast samples from January 2018 to December 2019 in Department of Breast Surgery, Liuzhou People’s Hospital, Guangxi Zhuang Autonomous Region were classified as cancer group, 30 cases of ductal atypical ductal hyperplasia (ADH) breast samples were classified as ADH group, and 30 cases of normal breast tissue were classified as normal group. Immunohistochemical staining was used to detect the positive expression of HPV16/18E6, TLR4, and NF-κB protein in three groups; the correlation between HPV16/18E6, TLR4, and NF-κB protein in cancer group and ADH group were analyzed; the relationship between positive expression of HPV16/18E6, TLR4, and NF-κB protein and clinicopathological features in cancer group were compared. Results The positive expression rate of HPV16/18E6, TLR4, and NF-κB protein among three groups were highly significantly different (P < 0.01). The positive expression rate of HPV16/18E6 in cancer group was higher than that in ADH group and normal group (P < 0.017); the positive expression rate of TLR4 and NF-κB protein cancer group was higher than that in ADH group and normal group, and ADH group was higher than normal group (P < 0.017). In cancer group and ADH group, HPV16/18E6 were positively correlated with TLR4 and NF-κB protein (P < 0.01), and TLR4 was positively correlated with NF-κB protein (P < 0.01). The positive expression rate of HPV16/18E6, TLR4, and NF-κB protein were correlated with histological grade, tumor diameter, lymph node metastasis, and clinicopathological stage (P < 0.05). Conclusion High-risk HPV infection plays an important role in the development and progression of breast cancer in conjunction with TLR4 and NF-κB inflammatory factors.
