Effects of dietary fiber on PTEN / PI3K / Akt / NF-κB signaling pathway after cerebral ischemia in rats#br# |

Abstract Objective To investigate the effects of dietary fiber supplementation on PTEN / PI3K / AKT / NF-κB signaling pathway after cerebral ischemia in rats. Methods Twenty-four rats were randomly selected to make cerebral ischemia model by intracranial injection of endothelin. Four rats died after the model was made. The remaining 20 rats were divided into ischemia group and ischemia + dietary fiber group according to random number table method, with ten rats in each group. In addition, ten rats were randomly selected as sham operation group and intracranial injection of equal amount of normal saline was used as control. Ischemia group and sham operation group were given normal diet, ischemia + dietary fiber group was given dietary fiber. The volume of cerebral infarction in ischemic group and ischemic + dietary fiber group was detected, and the protein expression levels of p-PI3K, p-Akt, PTEN and NF-κB/P65 in the peripheral cortex of rats in each group were detected by Western blot. Results There was no significant difference in cerebral infarction volume between ischemic group and ischemic + dietary fiber group (P > 0.05). The protein expression levels of p-PI3K and p-Akt in the ischemic group were lower than those in the sham group, and the differences were statistically significant (P < 0.05); the protein expression levels of p-PI3K and p-Akt in the ischemic + dietary fiber group were higher than those in the ischemic group, and the differences were statistically significant (P < 0.05). The protein expression levels of PTEN and NF-κB/ P65 in the ischemic group were higher than those in the sham group (P < 0.05), and the protein expression levels of PTEN and NF-κB/P65 in the ischemic + dietary fiber group were lower than those in the ischemic group, the differences were statistically significant (P < 0.05). Conclusion After cerebral infarction, dietary fiber can up-regulate PI3K / Akt signal and inhibit NF-κB pathway activation.
