Influence of Yisui Shengxue Granules on mRNA expression of C/EBPα, GM-CSF, GM-CSFRα and MAFB in bone marrow cells of mice irradiated by 60Co-γ rays |

Abstract Objective To study the influence of Yisui Shengxue Granules on CCAAT/enhancer binding protein α (C/EBPα), granulocyte-macrophage colon stimulating factor (GM-CSF), granulocyte-macrophage colon stimulating factor receptor alpha (GM-CSFRα) and transcription factor MAFB mRNA expression in bone marrow cells of mice irradiated by 60Co-γ rays, and to explore its mechanism of promoting bone marrow hematopoiesis. Methods Seventy-eight male Kunming mice were randomly divided into normal control group, model group, granulocyte-colony stimulating factor (G-CSF) group, low-dose, middle-dose and high-dose Yisui Shengxue Granules group. Two days before modeling, mice in low-dose, middle-dose and high-dose Yisui Shengxue Granules group received 12, 6, 3 g/(kg·d) Yisui Shengxue Granules ig respectively. On modeling day except normal control group, mice were established by 60Co-γ ray in other five groups. The 2nd day after modeling, mice in G-CSF group received 30 μg/(kg·d) G-CSF by injection, mice in normal control group and model group received the same volume distilled water, mice in low-dose, middle-dose and high-dose Yisui Shengxue Granules group received the same way as before modeling, the course lasted 14 days. Bone marrow cells were counted. The mRNA expression of C/EBPα, GM-CSF, GM-CSFRα and MAFB in BM cells was analyzed by real-time fluorescent quantitative polymerase chain reaction. Results The data showed that, Yisui Shengxue Granules could increase the count of bone marrow cells of mice irradiated by 60Co-γ rays (all P < 0.05), and up-regulate the mRNA expression of GM-CSF, GM-CSFRα and MAFB in BM cells (all P < 0.05). The effect of low-dose Yisui Shengxue Granules group was the best. Conclusion These results suggest that Yisui Shengxue Granules can promote bone marrow hematopoiesis, the mechanism may have some relation to up-regulating the mRNA expression of GM-CSF, GM-CSFRα and MAFB in BM cells.
