Abstract Neural precursor cell expressed developmentally down-regulated protein 4-2 (Nedd4-2) is a ubiquitin protein ligase of the E3 HECT family, which is known to bind and regulate a number of membrane proteins, and mediate diverse extracellular signals, such as cell differentiation and apoptosis. Epithelial sodium channel (ENaC) is an integral membrane protein, plays a crucial role in Na+ transport and fluid reabsorption in the kidney and lung, whose structural abnormalities or disfunctions may cause some diseases. At present, Nedd4-2 protein participate in the regulation of ENaC and regulate the function of the body. This paper reviewed the structure and function of the Nedd4-2 and ENaC, and summarized the regulation of ENaC trafficking by Nedd4-2.
