Anti-oxidative damage effect on PC12 cells transfected with human ApoE3 gene |

Abstract Objective To observe the effect of human ApoE3 gene high expression on the apoptosis of rat adrenal pheochromocytoma PC12 cells. Methods The pEGFP plasmid containing human ApoE3 gene and pEGFP plasmid were transfected with PC12 cells, and PC12 cell line was established that could stably express human ApoE3 gene and pEGFP plasmid through continuous screening. PC12 cells were allocated into 4 groups: normal control group, H2O2 group (cultured with 400 μmol/L H2O2 for 4 hours), liposome group (pEGFP transfection and H2O2 culture) and ApoE3 group (ApoE3 transfection and H2O2 culture). Cell survival was tested by CCK-8 method, cell apoptosis was detected by Hochest33258. Results Compared with the normal control group, cell survival rate was decreased in the other three groups (P < 0.05), while cell apoptotic was increased. The differences in the indexes mentioned above were not significant between H2O2 group and liposome group(P > 0.05). The cell survival rate was higher in the ApoE3 group, compared with H2O2 group and H2O2+liposome group (P < 0.05); the apoptosis was obviously lower in the ApoE3 group, compared with the H2O2 group and H2O2+liposome group. Conclusion The overexpression of ApoE3 gene has a great protective effect on the oxidative damage of PC12 cell mediated by H2O2.
