The effect of CD151 on the changes of Wnt signaling pathway in the combined targeting therapy of colorectal cancer |

Abstract Objective To explore the effect of CD151 on the changes of Wnt signaling pathway in colon cancer. Methods Western blot method was used to detect colon cancer cells (HT29) and CD151 gene knockout colon cancer cells (CD151--HT29) in CD151, LRG-1 and LGR-5 protein expression; the expression of LRG-1 HT29, CD151--HT29 cells and LGR-5 gene were detected by real-time quantitative PCR technique; the invasion ability of HT29, CD151--HT29 cells was inspected. Results The results showed that there was CD151, LRG-1 and LGR-5 protein expression in HT29 cells, but no CD151 protein expression in CD151--HT29 cells, and significantly decrease could be observed in the expression of LRG-1 and LGR-5 protein; real-time quantitative PCR results showe that, compared with HT29 cells, the expression of LRG-1 and LGR-5 gene in CD151--HT29 cells decreased significantly (P < 0.05). Cell invasion assay showed that HT29 cell invasion ability was stronger than that in CD151--HT29 cells (P < 0.05). Conclusion The expression of CD151 gene deletion can inhibit Wnt signaling initiation protein LRG-1 and LGR-5, and decrease the cancer cell invasion, which indicates that the CD151 gene has some role in the regulation of Wnt signaling pathway, to provide some scientific guidance for the colorectal cancer combined with targeted therapy.
