Abstract  Objective To explore the correlation between disintegrin-metalloproteinase 33 (ADAM33) gene S2, transforming growth factor-β1 (TGF-β1), and extracellular signal-regulated kinase (ERK) levels in Han nationality children with bronchial asthma in Urumqi. The level provides new ideas and targets for the prevention and treatment of asthma. Methods Outpatients and admissions of the Fifth Affiliated Hospital and First Affiliated Hospital of Xinjiang Medical University from July 2018 to July 2019 were selected, a total of 78 children with bronchial asthma in the Han nationality aged 3 to 14 were selected as the asthma group, and the same period when the preventive health care department was selected 45 cases of non-asthmatic children of Han nationality were regarded as non-asthmatic group. The ADAM33 S2 was typed by polymerase chain reaction, and the levels of IgE, TGF-β1 and ERK were detected by enzyme-linked immunosorbent assay. The differences of genotype and allele frequency between asthma group and non-asthma group were compared, and their correlation were analyzed. Results Comparison of ADAM33 S2 genotype and allele frequency between the two groups was statistically significant (P < 0.05). The levels of IgE, TGF-β1, and ERK in the non-asthmatic group were lower than those in the asthma group, and the differences were statistically significant (P < 0.05 or P < 0.01). TGF-β1 was positively correlated with ADAM33 S2 and ERK (r > 0, P < 0.05), and ADAM33S gene S2 was not correlated with ERK (P > 0.05). Conclusion One of the susceptibility genes in Han nationality children with asthma in Urumqi may be ADAM33 gene S2. The indicators of IgE, TGF-β1 and ERK in the asthma group are higher than those in the non-asthma group. TGF-β1 may up-regulate the expression of ADAM33 gene S2 in Han nationality children with asthma. ERK may up-regulate the secretion of TGF-β1.
